Shank2 is an excitatory postsynaptic scaffolding protein implicated in synapse regulation as well as psychiatric disorders including autism spectrum disorders. Conventional Shank2-mutant ($Shank2^{–/–}$) mice display several autistic-like behaviors, including social deficits, repetitive behaviors, hyperactivity, and anxiety-like behaviors. However, cell type-specific contributions to these behaviors have remained largely unclear. Here we deleted Shank2 in specific cell types, and found that male mice lacking Shank2 in excitatory neurons (CaMKII-Cre;$Shank2^{fl/fl}$) show social-interaction deficits and mild social-communication deficits, hyperactivity, and anxiety-like behaviors. Distinctly, male mice lacking Shank2 in GABAergic inhibitory neurons (Viaat-Cre;$Shank2^{fl/fl}$) display social-communication deficits, repetitive self-grooming, and mild hyperactivity. These behavioral changes were associated with distinct changes in hippocampal and striatal synaptic transmission in the two mouse lines. These results indicate that cell type-specific deletions of Shank2 in mice lead to differential synaptic and behavioral abnormalities.
Shank2는 흥분성 골격단백질로 흥분성 시냅스 조절을 담당하며, 자폐스펙트럼장애를 비롯한 정신질환에도 연관되어있다. Shank2 결손 생쥐는 사회성 결여, 반복 행동, 과잉행동 및 불안 증세와 같은 자폐 증상과 유사한 행동들을 보였다. 그러나 특정 세포 유형에서 Shank2가 위와 같은 행동들에 기여하는지는 아직 밝혀지지 않았다. 본 연구에서는 특정 세포유형에서 Shank2를 제한적으로 결손시킨 생쥐들을 만들었다. 흥분성 세포에서만 제한적으로 Shank2를 결손시킨 수컷 생쥐에서는 사회성 결여, 약간의 초음파 발성 및 소통 이상, 과잉행동 및 반복 행동 증세를 보였다. 다른 세포 유형인 억제성 세포에서만 제한적으로 Shank2가 결손된 수컷 생쥐에서는 초음파 발성 및 소통 결여, 반복적인 털 손질 행위와 약간의 과잉행동을 보였다. 이러한 행동 변화와 함께 해마와 선조체에서의 시냅스 전달 변화는 두 종류의 결손 생쥐에서 뚜렷이 구별되어 나타났다. 본 결과들은 세포 유형에 따른 Shank2의 제한적 결손이 뚜렷이 구별되는 시냅스 및 행동 이상 차이를 이끌었음을 시사한다.
