Most of nanoparticles for cancer therapy is cleared in liver without reaching target cells because of phagocytosis of macrophages. Attention to nanoparticles which are not uptaken by macrophages has been uprising to overcome this limitation. However, still large proportion of injected nanoparticles is cleared by macrophages, and complete avoidance of macrophage is very challenging. In this study, we researched the possibility to compensate the efficiency of delivery inducing macrophages to secrete uptaken cargo of nanoparticle, packaged in exosomes. We tried to increase the amount of re-excreted cargo by adding ceramide family lipid, which is known to induce cells to secrete exosomes, to nanoparticle liposome composition. We showed that ceramide family lipid contained in liposome enhanced the exosome secretion of macrophage and increased the amount of liposome cargo in exosome. Furthermore, we also showed that the increase of secreted material via exosomes can result in the enhancement of delivery to tumor cells.
암을 치료하기 위해 사용되는 나노 입자들은 대식세포의 식세포작용 때문에 표적 세포에 도달하지도 못한 채 간에서 배출되는 것이 대부분이다. 이를 극복하기 위해 대식세포에 흡수되지 않는 나노 입자에 대한 관심이 증가하고 있다. 그러나 여전히 많은 비율의 주입된 나노 입자가 대식세포에 의해 정리되며 대식세포의 흡수를 완전히 막는 것은 매우 어렵다. 본 연구에서는 대식세포에 흡수된 나노 입자의 수하물을 엑소좀의 형태로 다시 분비하게 만들어 전달 효과를 보완하는 가능성을 연구해보았다. 나노 입자의 한 종류인 리포좀의 구성성분에 세포의 엑소좀 분비를 유도한다고 알려진 세라마이드 계열의 지질을 추가함으로써 대식세포가 다시 분비하는 양을 늘리고자 하였다. 실험을 통해 리포좀에 포함된 세라마이드 계열의 지질이 대식세포의 엑소좀 분비를 증가시켰고 엑소좀 안의 수하물의 양 역시 증가시켰음을 확인하였다. 나아가 물질의 엑소좀을 통한 분비량의 증가가 종양 세포로의 전달량 증가로 이어질 수 있음을 확인하였다.
