NGL-2/Lrrc4 is one of the postsynaptic adhesion molecules that contains leucine rich repeats (LRRs), a shared feature of LRR proteins such as LTRRM and SALM. NGL-2 intracellularly interacts with the postsynaptic scaffolding protein PSD-95 and trans-synaptically binds to the presynaptic adhesion molecule Netrin-G2. Previous studies have reported that NGL-2 is expressed in a layer-specific manner in the brain and regulates diverse neurophysiological functions including excitatory synapse development and synaptic transmission. Moreover, recent bioinformatics studies of neuropsychiatric disorders such as autism spectrum disorder (ASD) and schizophrenia have strongly associated mutations of NGL2/LRRC4 in these diseases through approaches including whole exome and genome sequencing. The function of NGL-2, however, remains largely unknown from the behavioral and physiological perspectives. Here, my study show that NGL-2 regulates NMDA receptor-dependent synaptic transmission and plasticity and social and repetitive behaviors based on the data from Ngl$2^{-/-}$ mice that lack NGL-2 proteins. The defects in social interaction and repetitive behaviors, two core symptoms of ASD, were successfully rescued by the partial NMDA receptor agonist D-cycloserine. In addition, D-cycloserine improved abnormal NMDAR function and synaptic plasticity in these mice. Together, my study has found novel functions of NGL-2 in regulating NMDAR receptors, a core component of excitatory synaptic transmission, and critical brain function including sociability, cognition, and learning and memory. This study also highlights the importance of NGL-2 in neuropsychiatric disorders, and suggests abnormal NMDAR function might represent a therapeutic target for related disorders.
NGL-2는 류신영역이 주를 이루고 있는 시냅스후의 접착 단백질 중 하나로 잘 알려져 있다. 시냅스후의 세포내에서는 시냅스후의 주요 골격 단백질 PSD-95와 상호작용 하는 것으로 알려져 있으며 시냅스이전의 파트너 단백질 Netrin-G2와 트란스로 접촉하는 것으로 알려져 있다. 이전의 연구들에 의하면 NGL-2는 주로 흥분성 시냅스의 생성 및 여러 기능들을 조절하는 것으로 알려져 있으며 최근의 유전체학이나 생물정보학적 연구들에 의하면 자폐 스펙트럼 장애나 정신분열질환 같은 정신질환의 환자에서도 연관이 있은 것으로 보고가 되었다. NGL-2가 동물의 행동학적 그리고 생리학적 기능에 어떻게 관여하는지에 대한 연구는 아직까지 많이 진행이 되지 않은 상황이며 이 연구에서는 NGL-2가 NMDA수용체 의존적 시냅스 가변성 조절에 관여하며 사회성, 반복적행동 그리고 행동 학습에 주요 역할을 하는 것을 밝혀내었다. NGL-2 결손쥐에서는 이러한 행동학적 그리고 생리학적 장애를 유발하며 비정상적 행동 장애는 사람의 자폐 질환과 매우 유사한 증상을 보이는 것을 알 수 있었다. NMDA 수용체의 작용제 D-cycloserine을 처리함으로써 행동 장애와 생리학적 장애는 회복이 될 수 있는 것을 보여주고 있다. 이 연구는 NGL-2의 결손이 사회성 반복적행동의 장애를 동반한 자폐 스펙트럼 장애와 유사한 증상을 보이는 것을 처음 밝혀내었으며 시냅스 가소성에도 주요 역할을 하는 것을 밝혀내었다. NMDA 수용체의 작용제를 통한 회복 실험을 보여줌으로써 향후 정신질환 질병의 원인 연구의 새로운 가능성 중 하나를 제시하고 있다.
