Isolating actively proliferating cardioblasts is the first crucial step for cardiac regeneration through cell implantation. However, the origin and identity of putative cardioblasts are still unclear. Here, I uncover a novel class of cardiac lineage cells, $PDGFRα^+ Flk1^-$ cardioblasts (PCBs), from mouse and human pluripotent stem cells induced using CsAYTE, a combination of the small molecules Cyclosporin A, the rho-associated coiled-coil kinase inhibitor Y27632, the antioxidant Trolox, and the ALK5 inhibitor EW7197.
This novel population of actively proliferating cells is cardiac lineage-committed but in a morphologically and functionally immature state compared to mature cardiomyocytes. Most important, most of CsAYTE-induced PCBs spontaneously differentiated into functional $αMHC^+$ cardiomyocytes ($M^+CMs$) and could be a potential cellular resource for cardiac regeneration. Indeed, not only PCB-derived $M^+CMs$ but also undifferentiated PCBs effectively restored cardiac systolic function in a murine model of acute myocardial infarction through proper integration into the host myocardium.
활발하게 증식하는 심장근아세포들을 분리해내는 것은 세포이식을 통한 심장재생에 있어 가장 중요한 첫번째 단계이다. 하지만, 심장근아세포로 추정되는 세포들의 기원과 정체에 대해서 아직 잘 밝혀져 있지 않다. 본 연구에서는 네가지 화합물(Cyclosporin A, Y27632, Trolox, EW7197)을 이용하여 마우스와 인간줄기세포로부터 새로운 종류의 심장근아세포를 발견 및 생성하였다.
이 새로운 세포군집은 활발이 증식하며, 완전히 분화된 심장근세포와 비교하여 형태적 및 기능적으로 아직 미분화된 상태라는 것을 확인하였고, 네가지 화합물로부터 유도된 새로운 심장근아세포들은 자발적으로 기능을 갖춘 심장근세포로 분화하는 것을 확인하였다. 또한 네가지 화합물로부터 유도된 심장근아세포로부터 분화하여 얻은 심장근세포와 아직 분화되는 않은 심장근아세포 모두 마우스 심근경색 모델에서 심장기능을 회복시키는 것을 확인하였다. 이 연구에서 가장 중요한 것은, 네가지 화합물로부터 유도된 새로운 심장근아세포들이 자발적으로 기능을 갖춘 심장근세포로 분화되며, 심장재생에 있어서 굉장히 유망한 세포 자원이 될 수 있다는 것이다.
