Tissue engineering has been developed for improving or replacing biological tissue by manipulating the cells in vitro. In the process of transplanting the cells cultured in vitro to the patient again, there are difficulties such that the foreign substance enters the body and causes an unexpected immune response or toxicity in the process of decomposition, and the enzyme reaction can damage cell surface proteins or extracellular matrix proteins. To solve these problems, the cell sheet engineering has been developed, in which cells are constructed themselves without scaffold. Currently, the most used material in cell sheet engineering is a temperature sensitive polymer called pNIPAAm, which uses phase shift of polymer with temperature change. However, in the process of detaching the sheet by lowering the temperature, the cells are exposed to a low temperature. In addition, it takes a long time to form the cell sheet. In this study, two types of monomers with different surface energies were blended to control the surface free energy and to form an intermediate cell attachment state. In this situation, the divalent cations in the culture solution were depleted, and then cell sheet could be fabricated by weakening ligand binding of adhesion proteins in cell surface. By controlling the adhesive force between the cells and the culture surface, the cell sheet could be formed in a short time without changing the temperature for various cell types. Through this study, it is expected to manufacture various tissue-customized cell sheets production.
조직 공학은 생물학적 조직을 개선하거나 대체하기 위한 기술로서 발전되어 왔다. 그러나 체외에서 조작한 세포를 다시 환자에게 이식하는 과정에서, 외부 물질이 체내에 들어가 면역 반응을 일으키거나, 분해되는 과정에서 독성을 일으키는 등의 문제점, 체외에서 증식한 세포를 다시 수거하는 과정에서 효소 반응에 의해 세포 표면 단백질이나 세포 외 기질 단백질에 손상을 받는다는 문제점 등이 제기 되어 왔다. 이러한 문제점을 해결하기 위해, 지지 물질을 사용하지 않고, 세포 자체로 구조체를 만들어 이식하는 세포 시트 공학이 개발되었다. 현재 세포 시트 공학에서 가장 활발하게 사용되고 있는 물질은 pNIPAAm이라는 온도 감응 형 고분자로서, 온도 변화에 의한 고분자의 상 변이를 세포 시트 공학에 이용한다. 하지만 온도를 낮춰 시트를 떼어 내는 과정에서 세포가 낮은 온도에 노출되게 되며, 시트로 형성되기까지의 시간이 오래 걸린다는 단점이 있다. 본 연구에서는 표면 에너지가 다른 두 가지의 단량체를 혼합하여 표면 자유 에너지를 조절, 중간 수준의 세포 부착 상태를 만드는 방식과, 이러한 상황에서 배양액에 있는 2가 양이온을 제거하여 세포 표면에 있는 부착 단백질의 리간드 결합을 약화하는 방식을 결합하여 세포 시트를 제작 가능했다. 세포와 배양 표면의 접착력 조절을 통해 다양한 세포 종에 대해 온도 변화 없이 빠른 시간 내에 세포 시트를 형성할 수 있었고, 이를 통해 다양한 조직 맞춤형 세포 시트를 제작하거나 환자로부터 직접 얻은 세포를 활용한 세포 시트 제작 등에 적용 가능할 것으로 기대된다.
