Learning new information and storing memories are two of the most important functions of the brain. The purpose of the thesis is to develop a computational model that accounts for the mechanism of memory formation and maintenance. To approach this question, we designed three simulations covering the circuit, molecular, and system levels, respectively.
First, we discuss the roles of diverse spike-timing-dependent plasticity (STDP) profiles in flexible and stable memory formation. Through computer simulation, we found that an asymmetric learning rate generates flexible memory that is volatile and easily overwritten by newly appended information. Moreover, a symmetric learning rate generates stable memory that can coexist with newly appended information. Our results demonstrate that various attributes of memory functions may originate from differences in synaptic stability.
Second, we suggested the roles of intrinsic excitability in memory allocation. Using the model network, we successfully generated experimental results that showed that a neuron excited just before learning has a higher probability of being allocated in a memory ensemble than control group neurons. This result suggests that temporal excitability fluctuation not only can link two memories encoded close in time, but also boost the capacity of the network.
Finally, we present a novel systematic model in which the brain regions responsible for learning and coding are separated. We found that our model can account for the observed dynamics of an ensemble in the basolateral amygdala during fear conditioning. The simulated ensemble dynamics well described the observed data such as bi-directional plasticity, which never can be achieved by a single-layered network simulation.
Overall, our thesis presents an integrated approach to memory formation and maintenance through computer simulation. We hope this study can provide a unified description of how and why our memories are formed and can be maintained.
본 논문은 기억 저장과 유지의 회로 레벨 메커니즘을 설명하는 계산적 모델을 만드는 것을 목적으로 한다. 첫 번째 장에서, 우리는 다양한 형태의 스파이크 신호의 타이밍에 따른 가소성이 유연하거나 안정적인 기억의 형성에 미치는 영향에 대해 탐구하였다. 컴퓨터 시뮬레이션을 통해, 비대칭적 학습율에 의해 형성되는 기억은 휘발성이 높고 대칭적 학습율에 의해 생성된 기억은 안정적이라는 것을 보였다. 두 번째로, 신경 흥분성이 기억 할당에 미치는 영향에 대해 탐구하였다. 우리는 학습 직전에 흥분한 뉴런이 기억 앙상블에 포함될 확률이 높다는 실험 결과를 잘 재현할 수 있었고, 이 결과에 기반하여 신경 흥분성 변동이 가까운 기억을 연결시킬 뿐 만이 아니라 효율적인 기억 할당을 통해 신경망의 저장 용량을 늘릴 수 있다고 예측하였다. 마지막으로, 세 번째 장에서 학습과 부호화를 담당하는 뇌 영역이 분리되어 있다는 새로운 모델을 제시하였다. 이 모델을 통해 공포 기억 형성 과정 동안 편도체에서 관찰되는 앙상블 동역학을 설명할 수 있었다.
종합적으로, 본 연구는 기억의 형성과 유지에 대하여 시뮬레이션을 통해 통합적인 접근을 시도하였다. 본 연구는 회로, 분자적, 시스템 수준을 아우르는 통합적인 기억 모델을 제시한다는 점에서 큰 의의가 있다.
