Lactic acid bacteria (LAB) are a group of Gram-positive bacteria that produces lactic acid as a final end product in fermentation. Traditionally, LAB have long been used in the production of fermented dairy, meat and vegetable products as well as in wine and sourdough production. In addition, some species of LAB can produce health-related molecules such as antimicrobial peptides or bacteriocins that are used as biopreservative agents in foods. Recently, LAB have attracted attentions as a promising microbial cell factory and as mucosal delivery vehicles. Leuconostoc belongs to LAB is a non-sporulating, low G+C content, and a hetero-fermentative bacteria. Leuconostoc also plays important role in fermented food industry and has a broad spectrum of product such as lactic acid, alcohol, and aromatic compound. However, despite its importance in food and biotechnology industries, there has been little effort to develop genetic tools for engineering of the bacteria. In this study, I tried to engineer the expression vector system for enhancement of gene expression in L. citreum. For this purpose, I introduced bicistronic design (BCD) expression system into L. citreum. After the expression of target gene was observed in this expression system, Shine-Dalgarno (SD) sequence in the plasmid was engineered. SD2 library was constructed using super-folder green fluorescent protein (sfGFP) as a reporter, and highly fluorescent clones were isolated by FACS screening. The improvement of gene expression by the isolated strong SD2 was demonstrated with three recombinant proteins. Next, synthetic promoter library derived $P_{710}$ promoter was constructed in the engineered plasmid with strong SD2. The synthetic promoter library that induces sfGFP was screened by FACS. As a result, I could successfully isolate strong promoter and verify the strong promoter with two recombinant proteins. The expression system engineered with the promoter and SD2 showed about 1.6 times higher $\alpha$-amylase productivity than the previous expression system. Therefore, the expression vector system developed in this study will be useful for engineering of L. citreum in the future.
젖산균 (lactic acid bacteria) 은 발효에서 최종 최종 생성물로 젖산을 생산하는 그람 양성균이다. 전통적으로 젖산균은 발효 유제품, 육류 및 채소 제품의 생산에 오랫동안 사용되어 왔다. 또한 일부 젖산균 종은 항균 펩타이드나 박테리오신과 같은 인간의 건강에 이로움을 주는 물질을 생산할 수 있으며 이는 식품에서 생물학적 보존제로 사용된다. 최근 젖산균은 유망한 미생물 세포 공장 및 인체내 점막으로 다양한 물질을 전달 할 수 있는 매개체로 주목을 받고 있다. 류코노스톡은 이러한 젖산균에 속하며 포자를 형성하지 않고 낮은 G + C 함량 및 이종 발효 균주이다. 류코노스톡 또한 발효 식품 산업에서 중요한 역할을 하며 젖산, 알콜 및 방향족 화합물과 같은 광범위한 물질을 생산 할 수 있다. 그러나 이러한 식품 및 생명 공학 산업에서의 중요성에도 불구하고 류코노스톡에서 이용 가능한 유전적 도구들의 개발은 거의 없었다. 따라서 본 연구에서는 류코노스톡 시트륨에서 유전자 발현을 효율적으로 시키기 위한 발현 벡터를 개량하고자 하였다. 이를 위해 류코노스톡 시트륨에 bicistronic design (BCD) 발현 시스템을 도입했고 이 발현 시스템의 작동 여부를 확인 한 후, 발현 벡터에 존재하는 Shine-Dalgarno (SD) 서열이 조작되었다. SD2 라이브러리는 리포터로서 녹색 형광 단백질 (sfGFP)을 사용하여 제작되었으며, FACS 스크리닝으로 높은 형광 클론을 스크리닝 하였다. 스크리닝 된 강한 SD2에 의한 유전자 발현의 향상은 3개의 재조합 단백질의 생산에 적용함으로써 증명되었다. 다음으로, $P_{710}$ 프로모터 기반으로 합성 프로모터 라이브러리를 구축하였고, 형광 단백질과 FACS를 이용하여 라이브러리를 스크리닝 하였다. 결과적으로 강력한 프로모터를 성공적으로 스크리닝 할 수 있었고 두 가지 재조합 단백질로 분리된 강력한 프로모터의 기능성을 검증 할 수 있었다. 프로모터 및 SD2가 개량된 새로운 발현 벡터 시스템은 이전의 발현 시스템보다 약 1.6 배 높은 알파 아밀라아제 생산성을 보였다. 따라서 이 연구에서 개발 된 발현 벡터 시스템은 향후 류코노스톡이 산업용 균주로 개량하는데 유용하게 이용 되어 질 수 있을 것이다.
