The separation of proteins using Reverse Micelles in spray column was investigated. In this experiment, Extracellular Lipase and Intracellular Uricase were separated from aqueous solution. During the operation, the temperature was maintained at 27℃. For the deternimation of protein concentration, BCA micro assay was used. The amount of water in organic phase was assayed by Karl-Fisher Titrimeter.
We determimed the basis factors affecting the separation for extracellular and intracellular proteins such as pH of the aqueous phase, concentration of salt, surfactant and reducing agent mixing chamber.
The effect of pH was almost the same as the result of others for Lipase and Uricase. The optimum concentraton of salt for the separation was determined. As the concentration of surfactant increases, two maxima were shown in intracellular type. The amount of proteins in organic phase for intracellular type was increased with an increase in reducing agent.
Also the separation in spray column was performed at determined conditions. We investigated the optimum operation conditions, especially good extraction capacity, energy reduction and easy phase separation. The flow rate was optimum near 0.1 ml/sec. According to the increase of the circulation, the amount of extraction was almost constnat. The increase of length in column had positive effect on the amount of extraction. The amount of proteins in orgainc phase was increased with an increment of number of hole in distributor.
