The structures of chemically synthesized funtional and nonfunctional signal peptides of Escherichia coli ribose binding protein were determined by CD in trifluoroethanol solution which mimmics the amphiphilic environment. The estimated value of helix content of the wild type was found to be 98% at 25℃ and 86% 50℃. The estimated α-helix content of the mutant peptide was 55% at 25℃ and less than 10% at 50℃. It appeared that the Pro-9 residue in nonfunctional peptide disturbs the helix. Sine the α-helix segment of the wild type signal peptide is longer than that of the mutant signal sequence, it appears that the necessary condition for the translocation of RBP is the dimension of the helix in the signal sequence.
대장균 리보스결합 단백질내 신호서열의 돌연변이체와 야생형의 구조적인 차이를 CD를 사용하여 알아보았다. 야생형 신호서열 펩티드는 물/TFE 용액에서 총 24개 아미노산의 매우 안정된 helix 구조를 이루고 있었다. 한편 돌연변이체는 물/TFE 용액에서 총 11개의 아미노산이 helix를 이루는데 그 안 정도는 야생형에 비해 떨어진다. 이러한 2차구조 길이의 차이는 돌연변이체가 루이신-9 대신 프로린으로 치환되어서 helix형성을 방해하기 때문으로 보인다. 따라서 신호서열의 생리적 기능은 신호서열내 helix 길이에 의해 크게 좌우됨을 알수 있었다.
