Two members of mouse heat shock protein 70 family (M.W. 70 kDa) were investigated for the possible induction by bovine papillomavirus DNA transfection. One gene is expressed in male germ line as developmental stage specific manner (hsc 70), and the other is a heat-inducible hsp 70 gene, expressed in nearly all cell types. The promoter regions of these genes were isolated and linked to promoter-less CAT gene. CAT-recombinant DNAs were transfected to mouse fibroblast cells, and the CAT activities were analyzed.
By the heat shock, hsc 70 promoter showed two fold increased CAT activity as compared to normal condition. When BPV DNAs were cotransfected, CAT activity was also increased by two fold after 24 hrs. This increased CAT activity was also shown when E2 trans-activator gene of BPV was cotransfected. Heat-inducible hsp 70 promoter showed higher CAT activity than hsc 70 promoter in normal condition, however, transcriptional activity of hsp 70 promoter was not affected by heat shock or BPV DNA transfection.
분자량 70 kDa 인 쥐의 고온 충격 단백질들 중 두 종류의 단백질이 Bovine Papillomavirus DNA가 세포내로 유입 되었을 때 유도되는지 조사되었다. 한 단백질은 male germ line에서 발현되는 것이며 (hsc 70), 다른 한가지는 거의 모든 세포에서 고온 충격에 의해 유도된다 (hsp 70). 두 단백질 유전자의 promoter는 각각 분리되어, promoter를 가지고 있지 않은 CAT (chloramphenicol acetyl transferase) 유전자에 연결되어 쥐의 섬유 세포인 C127세포 내로 transfection되었으며, 세포 내에서의 CAT 활성도가 조사되었다.
고온 충격을 주었을 때 hsc 70은 정상상태에서 보다 2배 정도 늘어난 CAT 활성도를 보였다. BPV의 DNA가 세포 내로 유입 된지 24시간이 지난 후에 CAT 활성도가 고온충격을 주었을 경우처럼 2배 가량 증가한다. 이러한 CAT 활성도의 증가는 BPV의 E2 transactivator유전자를 세포내로 넣어주었을 때에도 나타난다. 고온 충격에 의해 유도되는 hsp 70은 고온 충격을 주지 않았을 경우에도 높은 CAT활성도를 보였으며, 고온 충격을 주었을 때나 BPV의 DNA가 세포 내로 유입되었을 때에도 비슷한 정도의 CAT 활성도를 보였다.
