An enhancement of the oxygen transfer rate in a 1 L bioreactor for mammalian cell culture by using a silicone rubber tubing as an oxygenator and the effects of the controlled dissolved oxygen(DO) concentration on the growth kinetics of Vero-6 cell were investigated. When the silicone membrane was used to supply oxygen to the culture broth, the oxygen transfer coefficients($K_La$) measured in deionized distilled water were markedly increased. Effect of surface aeration without the tubing aeration was very low under 1.0$hr^{-1}$ of $K_La$. The enhancing effects of agitation rates on $K_La$ were much more effective than aeration rates. The increase of $K_La$ with increasing tube length was observed as a results of the large surface area for oxygen supply. However, 2m of the tube length was adequate for a 1L vessel. The lager blade type of impeller was effective to enhance the $K_La$ values because of its high mixing intensity. In culture medium supplemented with 5% serum, $K_La$ values reduecd to approximately 40% probably due to the viscosity. Thus, oxygen enrichment in aeration and/or control of DO level were required for the adequate supply of oxygen to the cells. For this reason, a proportional control system of DO concentration in medium was developed by using a computer-coupled mass flow controller. The DO levels controlled by a microcomputer were very stable during the cultivation of Vero-6, while flow rates of air and/or oxygen enriched air were gradually changed depending on the DO concentration and the preset DO level. Growth of Vero-6 at the low level(10%) of DO was retarded, probably due to the oxygen limitation which was observed intuitively from the result of the high production of organic acid. Vero-6 cell cultured at higher DO than 30% could grow normally to the confluence on microcarriers.
산소 공급원으로 실리콘 튜브를 사용한 1 L 동물 세포 배양기에서 산소 전달 현상를 조사하였고 배양액의 용존 산소가 동물 세포 증식에 미치는 영향을 관찰하였다. 2차 증류수를 매질로 사용하여 표면통기가 유일한 산소공급원일 경우 $K_La$ 값이 1.0$hr^{-1}$ 이하로 매우 낮았고 이는 실제 동물 세포 배양에 적용했을 경우 산소 결핍 현상률 예상할 수 있었다. 그러나 2m의 실리콘 튜브를 사용하면 $K_La$ 값이 5.00 - 8.03 $hr^{-1}$ 6배 이상 증가하였다. 교반속도와 통기속도의 영향을 살펴보았는데 통기 속도에 비해 교반속도가 훨씬 효과적으로 $K_La$를 증가시켰다. 이는 반응기 내의 교반상태가 산소전달에 중요함을 나타낸다. 조사된 실리콘 튜브가 길수록 산소전달에 필요한 표면적이 넓어 산소 전달 속도는 증가하나 2m 가 효과적이었다. 교반기는 6 cm 길이의 pitched blade type 이 효과적이었다. 5% 혈청이 첨가된 배양배지에서는 혈청의 높은 점도 때문에 $K_La$ 값이 2m의 실리콘 튜브의 40% 수준으로 감소하였다. 배양액의 용존산소를 조절하기 위해 컴퓨터와 연결된 mass flow controller를 이용한 proportional control을 시도하였다. 부착성 동물 세포인 Vero-6 세포를 배양할 때 용존산소는 ±2% 이내로 매우 안정하였다. 용존산소를 50% 와 30% 로 조절하였을 경우 Vero-6 세포는 최고 세포 농도가 $1.5\times10^6$ cells/mL 와 $1.7\times10^6$ cells/mL 로 정상적인 증식을 보였으나 10% 로 조절하였을 경우 $3.8 \times10^5$ cells/mL로 세포 증식이 억제되었다. 이것은 산소 결핍 현상에 의한 결과라 사료된다.
