Regulatory T cells play an important role in maintenance of immune homeostasis and tolerance through regulation of excessive immune response. As well as suppressive potential but also migration ability of Treg cells is also a crucial factor that promotes their role. However, mechanism underlying this migration of Treg cells during acute viral infection has not been specifically studied. In this study, we focused on chemokine receptors that influence Treg cells to move to the target site where severe immune response occurring.
During lethal influenza infection, we observed increased frequency and absolute count of Treg cells in the lungs. To determine the chemokine receptors related to the appropriate localization of Treg cells, we analyzed various chemokine receptors on lung-localized Treg cells and CCR2, CCR5 and CXCR3 were chosen as candidates. Furthermore, we confirmed that three chemokine receptors were related to migration of Treg cells through performing in vitro migration assay and adoptive transfer experiment using chemokine receptor deficient mice.
During acute hepatitis A infection, expression of CCR5 was up-regulated on peripheral circulating Treg cells so we denoted them as candidates participating in migration of Treg cells to liver. In addition, the levels of chemokine related to CCR5 were higher than healthy controls in patients’ serum. Lastly, we observed that the highly expression of $CCR5^+$ on Treg cells located in liver during acute hepatitis A infection.
With our present study, we investigated for the first time the expression of various chemokine receptors involved in the migration of Treg cells during acute viral infection. We suggest that the expression of CCR2, CCR5 and CXCR3 has a critical role in localization of Treg cells into the flu-infected lungs. Furthermore, in case of acute hepatitis A virus infection, we purpose that CCR5 seems to participate in migration of Treg cells to the liver.
조절 T 세포는 우리 몸에서 자가항원에 대한 과도한 면역반응을 조절하여 면역체계의 항상성을 유지하는 데 중요한 역할을 한다. 조절 T 세포는 그들의 역할에 있어서 과도한 면역반응을 일으키는 세포들을 억제하는 기능뿐만 아니라 타겟 장소에 도달하는 이동능력 또한 중요하다. 본 연구에서는 이러한 조절 T 세포의 이동능력에 관하여 급성 바이러스 감염모델에서 케모카인 수용체 발현을 중심으로 연구를 진행하였다.
먼저 마우스 인플루엔자 감염 모델에서 우리는 바이러스의 감염 반응이 일어나는 폐에 위치한 조절 T 세포가 CCR2와 CXCR3, CCR5를 말초에 위치한 조절 T 세포에 비해 높게 발현되어 있는 것을 확인 하였다. 인플루엔자 감염 시에 각각의 수용체가 소실된 유전자변형마우스에서 분리한 조절 T 세포가 실제로 폐에 위치하는 데에 어려움이 있음을 입양전달실험을 통해 확인하였다.
인간의 급성 A형 간염 바이러스 감염모델에서는 말초혈액을 이용하여 조절 T 세포에 발현한 케모카인 수용체를 관찰하였을 때, CCR5가 정상인에 비해 환자에서 높게 발현되고 있는 것을 확인하였고, 같은 환자의 회복기에는 정상인과 같은 수준으로 감소하는 것을 알 수 있었다. 환자의 간에 위치한 조절 T 세포가 CCR5를 높게 발현하는 것을 확인함으로써, CCR5가 조절 T 세포의 이동에 중요한 역할을 한다는 것을 알 수 있었다
이러한 결과들을 종합하여 볼 때, 급성 바이러스 감염의 모델로 인플루엔자와 A형 간염 바이러스 감염 상황에서 조절 T 세포의 이동에 관련한 수용체들의 서로 다른 발현 패턴을 보이는 것을 관찰하였고, 이들이 조절 T 세포가 타겟 장소로 이동하는 데에 중요한 역할을 하는 것을 알 수 있었다. 본 연구의 결과가 과도한 자가면역반응 질환이나 효과 T 세포의 반응의 유도가 필요한 질환에서 조절 T 세포의 분포를 조절할 수 있는 치료법의 기반이 될 것으로 기대한다.
