During inflammation lymphatic vessels (LVs) are enlarged and their density is increased through the effects of pro-lymphangiogenic factors including vascular endothelial growth factors, which facilitate the migration of activated immune cells and antigens. After antigen clearance the expanded LVs shrink and activated immune cells undergo apoptotic cell death to maintain homeostasis. Several cytokines secreted from T helper type 1 (Th1) and Th2 cells have been discovered to have anti-lymphangiogenic roles in inflammatory conditions. However, less is known about the effects of cytokines from Th17 cells. Here, we show that interleukin (IL)-17A, secreted from Th17 cells, is a negative regulator of lymphangiogenesis in Th17-mediated immune responses, especially during the resolution phase of inflammation. Moreover, IL-17A suppresses expression of major lymphatic markers in lymphatic endothelial cells and decreases in vitro LV formation. To investigate the role of IL-17A in vivo, we utilized a cholera toxin-mediated Th17 inflammation model and identified inflammation and resolution phases based on the numbers of recruited immune cells. IL-17A, markedly produced by Th17 cells even after the peak of inflammation, was found to participate in the negative regulation of LV formation. Moreover, blockade of IL-17A resulted in not only increased density of LVs in tissues, but also their enhanced function. Taken together, these findings improve the current understanding of the relationship between LVs and inflammatory cytokines in pathologic conditions.
면역반응 중에 림프관은 다양한 림프관성장인자들의 영향으로 확장되고, 그 밀도가 높아진다. 확장된 림프관은 항원과 활성화된 면역세포들의 이동을 용이하게 한다. 하지만 항원들이 모두 제거된 후에 활성화된 면역세포들이 세포자살기전을 겪듯이, 확장되었던 림프관 역시 축소되어 항상성을 유지하는데 기여한다. 최근 Th1, Th2 세포들이 분비하는 사이토카인들이 림프관형성을 억제하는 기능을 한다는 사실이 밝혀졌다. 본 연구에서는 Th17 세포로부터 분비되는 사이토카인이 림프관형성에 미치는 영향을 조사하였다. 또한 IL-17A가 림프관 내피세포에서 림프관 특이적인 유전자 발현을 억제하고, in vitro 림프관 형성을 저해하는 것을 확인하였다. In vivo에서 IL-17A의 역할을 관찰하기 위해 콜레라 독소로 유도된 염증모델을 도입하였고, 염증부위로 이동한 면역세포들의 수를 바탕으로 염증기와 해소기를 구분하였다. 염증기 이후에도 Th17 세포에서 분비되는 IL-17A는 림프관형성억제에 참여하는 것으로 확인되었다. 또한 IL-17A를 차단했을 경우 림프관의 밀도가 높아졌을 뿐 아니라, 기능도 향상된 것을 확인했다. 이러한 결과들을 통합해볼 때, Th17 세포로 인한 염증반응의 해소기에서 Th17 세포로부터 분비되는 IL-17A가 림프관형성을 억제한다는 사실을 알 수 있었다. 이는 병리적인 상황에서 림프관과 염증 사이토카인의 관계에 대한 이해를 더욱 넓혀줄 것으로 기대된다.
