A tumor vasculature which are destabilized in structure and function leads to limited drug delivery, hypoxia, unfavorable tumor microenvironment, and even metastasis. I performed a side-by-side comparison of ABTAA (Ang2-Binding and Tie2-Activating Antibody) and ABA (Ang2-Blocking Antibody) in mice with subcutaneously implanted Lewis lung carcinoma, and with spontaneous mammary cancer. I found that Tie2 activation-induced tumor vascular normalization, leading to enhanced blood perfusion and chemotherapeutic drug delivery, and reduced tumor growth and metastasis. Moreover, ABTAA favorably altered the immune cell profile within tumors. Together, my findings establish that simultaneous Tie2 activation and Ang2 inhibition form a powerful therapeutic strategy to elicit a favorable tumor microenvironment and enhanced delivery of chemotherapeutic agent into tumors.
불안정한 암 혈관은 항암제 전달의 감소, 저산소증, 악화된 종양미세환경, 그리고 암 전이의 원인이 된다. 본 연구에서는 새롭게 개발된 항체신약의 항암효과를 두 가지의 마우스 종양모델 (LLC 모델, 유방암 모델)을 이용하여 확인하였다. 암 혈관의 Tie2 수용체가 항체신약에 의해 활성화 될 경우 암 혈관의 정상화가 일어나며 종양 내부로의 혈류증가, 항암제 전달증가, 종양성장 감소 및 전이의 감소가 일어남을 확인하였다. 더욱이 새로운 항체신약은 종양 내부의 미세환경을 변화시킴을 확인하였다. 종합적으로, 암 혈관의 Tie2 수용체 활성화에 의해 유도된 암혈관 정상화는 종양 내부의 미세환경을 변화시키고 항암제의 전달을 증가시킴으로써 효과적인 치료수단이 될 수 있을 것으로 기대된다.
