In drug development, high cost of drug discovery has been a major issue due to the large chemical space and low bioactivities of found compounds. In drug screening tests, human metabolites have been thoroughly used to test possible bioactivity and toxicity of compounds of interest. By expanding the usage of human metabolite information in current drug discovery field, we propose a novel method to predict drug compounds from natural products by using human metabolite similarity. To find human metabolites with therapeutic effects, we gathered known associated diseases and analyzed a gene network to cover possible therapeutic effects. Drug compounds were searched from natural products, for they are much similar to human metabolites with higher bioactivities than synthetic compounds are. The similarity between natural products and human metabolites is measured through structure, target and phenotype analysis, and the effect terms of human metabolites were mapped to the paired natural products. As a result, possible drug compounds were found in natural products by our method.
약품 개발 분야에 있어서 많은 양의 화합물 속에서 신약을 탐색하는 것은 시간적으로나 금전적으로 큰 문제가 되고 있다. 인체 대사물질은 약품 개발 과정에 있어서 해당 약물의 체내 활성도, 혹은 독성을 유추하기 위해 자주 사용됐는데, 본 연구에서는 기존에 사용되던 인체 대사물질 정보를 확장하여, 인체 대사물질과의 유사성 비교를 통해 천연물 중에서 약물 후보군을 예측하는 방법론을 소개한다. 먼저 치료 효과가 있는 인체 대사물질을 찾기 위해 대사물질과 연관된 기존에 알려진 질병 정보를 수집하였으며, 유전자 네트워크 분석을 통해 해당 대사물질과 관련된 치료 혹은 질병 정보를 추가로 수집하였다. 천연물의 경우 합성 약물보다 인체 대사물질과 비슷할뿐더러, 체내 활성도 또한 높아, 천연물을 대상으로 약물 후보군을 탐색하였다. 인체 대사물질과 천연물 간의 유사성은 구조, 작용 대상, 전반적인 효능 분석을 통해 이루어졌고, 서로 유사하다고 판단된 천연물은 연관된 인체 대사물질의 효능을 가질 것으로 예측하였다. 결과적으로 전체 방법론을 통해 약리 작용을 가질 것으로 예상되는 천연물을 추출하였다.
