Cells migrate by directing Rac1 and Cdc42 activities as well as actin polymerization towards their front. Previous studies proposed that this polarization process requires a local positive feed-back in the front involving Rac small GTPase and actin polymerization with PI3K likely playing a coor-dinating role. Here we show that PLEKHG3 is a PI3K-regulated RhoGEF for Rac1 and Cdc42 that se-lectively binds to newly polymerized actin in the front of migrating fibroblasts. Optogenetic inactiva-tion of PLEKHG3 showed that PLEKHG3 is indispensable not only for inducing but also for maintain-ing cell polarity. By selectively binding to newly polymerized actin, PLEKHG3 promotes local Rac1/ Cdc42 activation to induce more local actin polymerization which in turn promotes recruitment of more PLEKHG3 to induce and maintain frontness. Thus, autocatalytic reinforcement of PLEKHG3 localiza-tion to the front provides a molecular basis for the proposed positive feedback loop that is required for cell polarization and directed migration.
Small G 단백질은 세포의 이동 및 발생 등 아주 다양한 신호 전달 과정에서 중요한 분자스위치 역할을 하는 단백질이다. 따라서 지금까지 small G 단백질의 유전자 변이나 잘못된 조절 메커니즘에 대해 아주 많은 연구가 진행되었다. 세포의 정상적인 기능에서 중요한 역할을 하는 small G 단백질은 GEF, GAP, GDI 조절단백질에 의해 활성이 조절된다. 이 중 GEF 단백질은 small G 단백질을 활성화 시키는 역할을 하며, GEF 단백질의 비정상적인 활성 조절은 암 등 인간의 질병과 밀접하게 연관되어 있다. 인간 유전체 내에 GEF 단백질이 70여 개 이상이 존재함에도 불구하고 아직까지 조절 메커니즘이 정확하게 연구가 잘 되어 있지 않고, 20여 개의 Rho small G 단백질 중 Rac1, Cdc42, RhoA 세가지 Rho small G 단백질에만 연구가 집중되어 이루어지고 있다.
본 연구에서는 컨포컬 이미징을 통해 66개의 GEF의 세포 내 분포를 분석하여 세포이동 시 Rac1/Cdc42를 활성화하여 세포이동을 조절할 가능성이 높은 세포막, 액틴필라멘트 및 미세소관에 존재하는 GEF를 발굴하였다. 발굴된 GEF를 이용하여 세포이동을 활성화하는 GEF를 동정하였고, 그 중에서 PLEKHG3가 세포이동 시 세포의 leading edge로 활발하게 이동하는 것을 확인하였다. 지금까지의 세포이동 연구에서는 세포이동 시 PI3K를 통해 Rac1/Cdc42가 활성화되고 액틴중합이 일어나고, 이러한 새롭게 형성된 액틴필라멘트에 의해 다시 PI3K가 활성화되는 positive feedback 메커니즘이 알려져 있었다. 하지만 액틴필라멘트가 어떻게 다시 PI3K와 Rac1/Cdc42를 활성화하는지는 알 수가 없었다. 본 연구에서 찾은 PLEKHG3가 액틴 필라멘트에 아주 강하게 결합하는 새로운 단백질임을 알 수 있었고 세포가 이동할 때 형성되는 액틴필라멘트에 결합하여 Rac1/Cdc42을 활성화하고 이를 통해 다시 액틴 필라멘트가 활성화 된다는 새로운 positive feedback 메커니즘을 밝힐 수 있었다.
본 연구에서는 지금까지 그 기능이 전혀 알려지지 않았던 PLEKHG3의 세포이동 시 Rac1/Cdc42를 활성화하여 액틴 중합을 유도하고 동시에 액틴 필라멘트에 결합함으로써 Rac1/Cdc42을 활성화함을 알 수 있었다. 이 연구에서 RhoGEF-Rac1/Cdc42의 새로운 단백질의 조절관계에 대한 정보를 제공함으로써, 차후 특정 GEF와 small G 단백질의 조절을 인위적으로 제어하여 인간의 질병을 치료할 수 있는 유용한 데이터가 될 것이다.
