Acetylation of ε-amino groups of lysines changed the conformation of glycinin isolated from soybean. Soy glycinin with lysine residues modifications of 0%, 25%, 65% and 95% were used for the experiment. Surface hydrophobicity was found to increase in about 3 times when lysine residues were excessively modified to above 95%. Masking of charged lysine residues, exposure of hydrophobic interior and subunit dissociation should have contributed to the conformational change that can be detected by UV spectra and the change of the intensity of effectiveness of pH, salts, temperature and conglycinin aggregation or solubilization of glycinin. The effect of acetylation on solubility of protein was investigated. Solubility of acetylated glycinin was increased and isoelectric point was shifted to acidic region. Salts (NaCl, $CaCl_2$) were found to little affect solubility of acetylated glycinin in contrast to native one. Heat stability of acetylated glycinin was increased, conglycinin affected little on solubility of acetylated glycinin in contrast to native one.
콩의 주요 저장 단백질인 Glycinin의 라이신잔기를 적당량의 Acetic anhydride를 이용하여 25%, 65%, 90%, 95% 로 아세틸화 시켰다. 아세틸화에 의한 Glycinin의 구조적 변화를 UV 흡광법, 형광법에 의해 측정한 결과 Surface hydrophobicity가 증가하였다. 이같은 구조적 변화에 의해 기능성이 변하였다. 기능성의 지표로는 용해도를 사용하였는데, 아세틸화시 용해도가 증가하였고, 등전점이 산성쪽으로 이동하였다. Glycinin의 용해도 증가를 일으키는 NaCl은 아세틸화된 Glycinin 의 경우에는 큰 영향을 주지못했고, pH 6 이상에서 Glycinin의 침전을 일으키는 $CaCl_2$ 는 아세틸화된 경우에는 침전을 일으키지 못했다. 또 아세틸화시 열 안정성이 증가했으나, 열안정성에 대한 NaCl의 효과는 감소하였고 $CaCl_2$ 는 별 효과가 없었다. 아세틸화시 Conglycinin이 Glycinin 을 열안정화 시키는 효과는 감소하였다.
