The microorganisms grown on a minimal medium with aniline as a sole source of carbon, nitrogen, and energy were isolated, identified, and characterized.
Pseudomonas acidovorans strain 4Al harbored a plasmid of ca. 230 kilobases, shown to be involved in aniline metabolism.
The substrate specificity of aniline-dioxygenase from Pseudomonas testosteroni strain 6Fl was found to be restricted to native form of aniline, indicating that it did not act on aniline derivatives or cationic state of aniline. In addition, this enzyme might not be coordinated with other enzymes of the β-keto-adipate pathway. Utilization of aniline was promoted by the addition of other utilizable carbon sources to the bacterial cells of P. testosteroni strain 6Fl in lag phase and in starved condition. It was shown that the NADH level in the cells related to the aniline uptake rate.
아닐린(aniline)을 유일한 탄소, 질소, 그리고 에너지원으로 이용할 수 있는 미생물을 분리 동정하고, 그 특성을 조사하여 보았다. $\underline{Pseudomonas}$ $\underline{acidovorans}$ strain 4Al에 존재하는 plasmid는 크기가 약 230 kilobases로 아닐린 분해 효소 체계를 수록하고 있는 것으로 판단되었다.
$\underline{Pseudomonas}$ $\underline{testosteroni}$ strain 6Fl의 anilinedioxygenase는 기질특이성을 보이는 효소로서 아닐린 유도체나 양이온 형태의 아닐린에는 작용하지 않았다. 그리고, 이 효소는 β-keto-adipate pathway의 다른 효소들과는 다른 operon상에 존재하는 것으로 추정되었다.
P. $\underline{testosteroni}$ strain 6Fl 의 경우, 이용 가능한 다른 탄소원을 첨가해줌으로써 휴지기 또는 정지기 상태의 cells 들의 아닐린 이용속도를 증가시킬 수 있었다. 이러한 이용속도의 변화는 cell내의 NADH양과도 밀접히 관련되어 있는 것으로 판단되었다.
