Mitochondrial aspartate aminotransferase(m-AAT) is known as a special transport protein which goes into the matrix side of isolated mitochondria through the membrane in vitro.
In order to understand the transport mechanism of m-AAT, a pH dependent binding test has been performed. It was found that the binding of m-AAT was maximum at pH 4 but no appreciable binding was observed at neutral pH in the CL:PC 60% vesicles. The transport of m-AAT across the bilayer was not detected in all pH range studied.
The binding pattern of cytosolic aspartate aminotransferase was different from that of mitochondrial isozyme. A detectable amount of binding of cytosolic isozyme was observed at neutral pH.
The irreversible binding was observed only at pH 4, and the segment of the enzyme which penetrates into the bilayer was identified by hydrophobic labeling with dansyl chloride.
Mitochondrial aspartate aminotransferase 의 미토콘드리아 내부로의 통과 mechanism 을 이해하기 위해서 실험을 수행하였다. m-AAT의 pH에 따른 결합 양상은 pH 에 따라 큰 차이를 나타내었으며, CL:PC 60% (1:4) 의 인지질 막에서는 중성 pH 에서 결합이 되지 않았으며, pH 4 에서 최고로 결합되었다. 이러한 결합은 높은 이온 강도하 에서도 일부 유지 되었는데, pH 4의 경우에만 관찰되었다. 이러한 비가역적 결합에는 소수성 작용이 관여함을 나타낸다. 실험을 수행한 모든 pH 에서, m-AAT가 인지질막의 내부로 통과되는 것은 관찰되지 않았다.
Cytosolic aspartate aminotransferase 는 m-AAT 와는 다른 결합 양상을 나타내었으며, 중성 pH 에서도 어느 정도 결합 되었다.
