Swept-source optical coherence tomography (SS-OCT) is a noninvasive, high sensitivity, cross-sectional imaging modality that provides structural information of biological samples with high resolution. Additional functional imaging can be obtained through the application of Doppler techniques to image cerebral or retinal blood flow. However, SS-OCT requires high phase stability for Doppler imaging. In SS-OCT the measurement of interference fringes is not perfectly repeatable due to instabilities in the data-acquisition board (DAQ) and the wavelength swept-source laser which lead to SS-OCT suffer from phase unstability. In this research, phase-stabilized SS-OCT with high-speed (540 kHz) is developed with a robust and computationally efficient approach for phase stabilization. As a future work, the system will be used as a tool for studying blood flow and neurovascular coupling.
SS-OCT는 살아있는 생체 샘플에 대해 비침습적, 고해상도 이미징이 가능한 구조 이미징 기술이다. 이러한 구조 이미징 이외에도 SS-OCT에 도플러 이미징 기술을 접목하여 뇌나 망막에 존재하는 혈류 이미징이 가능하며 이를 위해 SS-OCT는 높은 위상 안정성이 요구된다. 하지만 일반적으로 SS-OCT는 파장변환레이저 즉, 광원이 불안정하여 고정되어 있는 동일한 샘플에 대해 매 레이저 반복시마다 서로 다른 간섭신호가 출력된다. 이로 인해 시스템의 위상이 불안정하고 따라서 이러한 일반적인 SS-OCT로는 정량적인 혈류 이미징이 불가능하다. 본 연구에서는 540 kHz의 고속 SS-OCT 시스템에 위상 안정화를 구현하여 정량적 혈류 측정이 가능하며 이를 통해 신경혈관연동 연구에 적용 가능한 시스템을 개발하였다.
