Angiopoietin-1 (Ang1), a potential growth factor for therapeutic angiogenesis and vascular stabilization, is known to specifically cluster and activate Tie2 in high oligomeric forms, which is a unique and essential process in this ligand-receptor interaction. However, highly oligomeric native Ang1 and Ang1 variants are difficult to produce, purify, and store in a stable and active form. To overcome these limitations, we developed a simple and active dimeric CMP-Ang1 by replacing the N-terminal of native Ang1 with the coiled-coil domain of cartilage matrix protein (CMP) bearing mutations in its cysteine residues. This dimeric CMP-Ang1 effectively increased the migration, survival, and tube formation of endothelial cells via Tie2 activation. Furthermore, dimeric CMP-Ang1 induced angiogenesis and sup-pressed vascular leakage in vivo. Despite its dimeric structure, the potencies of such Tie2-activation-induced effects were comparable to those of a previously engineered protein, COMP-Ang1. We also revealed that these effects of dimeric CMP-Ang1 were affected by specified N-glycosylation in its fibrinogen-like domain. Taken together, our results indicate that dimeric CMP-Ang1 is capable of activating Tie2 and stimulating angiogenesis in N-glycan dependent manner.
Angiopoietin-1 (Ang1)은 혈관내피세포에 특이적으로 발현하는 Tie2 수용체의 리간드로서, 혈관과 림프관의 성장을 조절하는 혈관세포성장인자이다. 또한 Ang1은 혈관내피세포의 생존율을 증가시키고 혈관의 누수를 차단하여 치료용 목적으로 사용 될 수 있는 가능성을 가진다. 하지만 Ang1의 특이한 분자구조적 특징으로 인해 대량생산 및 정제하기 어렵고 Ang1 활성의 불안정이 쉽게 유발된다는 문제점이 있다.
본 연구에서는 이러한 문제를 해결하기 위해 N-terminal을 cartilage matrix protein (CMP)의 coiled-coil 도메인으로 치환하고 cysteine를 alanine으로 치환하여 이중체 구조를 가진 CMP-Ang1을 개발하였다. 기존의 이중체 Ang1 variant들과 달리, 이중체 CMP-Ang1은 효과적으로 Tie 수용체 및 하위 신호전달까지 활성화시켰다. 그러나 다른 Ang1 variant들과 달리, 이중체 CMP-Ang1은 생산하는 세포주에 따라 활성이 달라짐을 확인하였다. LC/MS분석을 통해 1개의 N-glycan 자리가 있으며 생산 세포주에 따라서 이중체 CMP-Ang1에서로 다른 N-glycan이 형성되어있음을 확인하였다. 이러한 N-glycan은 Tie2 수용체와의 결합에는 영향을 주지 않지만 N-glycan의 종류에 따라 이중체 CMP-Ang1의 Tie2 수용체 활성이 달라진다는 것을 확인하였다.
이중체 CMP-Ang1은 효과적으로 혈관내피세포의 이동, 생존 및 튜브 형성을 증가시키고, in vivo 실험을 통해 새로운 혈관 형성을 증가시키고 혈관 누수를 줄이는 효과를 보였다. 이런 결과를 토대로 이중체 CMP-Ang1의 향후 차세대 혈관신생 촉진 치료제로서의 가능성을 제시하고 있다.
