In the recent years, interest in cell culture technique has been grown due to the need for the production of interferon, monoclonal antibodies, various hormons, vaccines for viruses, and other cell-secreted products which have a potential medical use. In the point of view, the microcarrier culture, in particular, for the growth of anchorage-dependent animal cells has been innovated and significantly improved.
We employed a microcarrier culture technique for MRC-5 cell (human embryonic lung cell) and GM-2504 cell (human foreskin fibroblast strain which is trisomic for chromosome 21). The major parameters which affected yields of cell from microcarrier (Cytodex 1) culture were determined.
Firstly, optimal concentrations of microcarrier were determined by growth curves. Also, the attachment of cells to beads was established within 4 hours in this system. It has been found that the more inoculation density of cells were, the higher final concentrations of cells were obtained. Cells per mlin microcarrier culture has increased three fold more than that of the standard culture (monolayer culture on petri plate).
Secondly, interferon yields produced from GM-2504 cells grown on microcarriers increased about two-fold using superinduction method than that for the standard culture. Heparin effect as polyanion materials was also applied to the microccrier culture in the production of interferon in this system. Heparin effect allowed to enhance about two-fold in interferon production more than normal superinduction methods of microcarrier culture.
Finally, we tested potential of collagen immobilized with succinic anhydride as inducers or/and coinducers of induction of interferon. None of them, however, induced positively the production of interferon in this system.
바이러스 증식과 암세포 증식을 억제하는 것으로 알려진 인터페론이란 단백질을 비롯하여 여러가지 바이러스 백신, 호르몬, 항원 등 의약용으로 사용되는 물질을 대량 생산하는데 있어 최근에 새로운 방법으로서 microcarrier 배양 방법이 발전해 왔다. 실험실에서 조직 배양되는 정상적인 동물세포는 반드시 배양용 용기나 접시의 바닥에 붙어 자라는 성질때문에 이러한 세포의 대량 증식은 붙어 자랄 수 있는 표면적에 의해 크게 제한을 받아왔다. 그러나 microcarrier라는 구슬모양의 작은 크기(지름: 160μm-230μm)의 beads는 거의 무제한의 표면적을 제공할 수 있으므로 조직배양에 혁신을 가져왔다.
이 배양 방법을 이용하여 본 실험에서는 세포를 종래의 petri plate 배양방법에서 보다도 3배 가량 증가시켰으며 또한 인터페론의 양도 약 2배 가량 증가시켰다. 이는 microcarrier배양에 영향을 미치는 여러 parameters 를 적절하게 조정해 줌으로써 가능했다. parameters중에서 가장 중요한 것으로 생각되는 것이 microcarrier 배양이 시작될 쯤에 접종시키는 세포의 밀도가 많을수록($1.26\times10^5 cells/ml$) 최종 세포 수율이 증가한다는 것을 알 수 있었다. 또한 beads의 농도와 배양계의 동질화에 영향을 미친다는 것을 알 수 있었다.
마지막으로 실험실에서 인터페론의 유도물질로서 합성된 숙신산 무수물이 고정화된 collagen은 인터페론 생성능력이 없는 것으로 밝혀졌다.
