The activation of human plasminogen to active fibrinolytic enzyme plasmin (EC 3.4.4.14) was studied with streptokinase.
The activation of human plasminogen to an active enzyme plasmin is occured via hydrolytic cleavage of Arginyl-Valyl endopeptide bond. Several forms of the plasminogen activator have been found many different source including humoral origin. Among them, an urinary activator, urokinase, has been studied mostly and thus used as a prototype activator for the biochemical mechanism of the plasminogen activation process. However, a series of study on the plasminogen activation by bacterial activator, streptokinase, from β-haemolytical pathogenic bacteria, indicates that its mode of action is quite different from that by urokinase.
In this communication, we report our findings on the nature of streptokinase as an allosteric protein requiring human plasminogen for the catalytic activity.
When the amount of streptokinase is fixed and the plasmingen concentration is varied, the concentration dependent rate profile showed a typical saturation curve. However, when the ratio of plasminogen and streptokinase was small, the rate profile becomes sigmoidal.
The kinetic data along with other results suggest that streptokinase forms a complex with plasminogen to become an active enzyme, but once bound with plasminogen it becomes an active enzyme.
SK에 의한 human plasminogen 의 활성화는 plasminogen 분자의 peptide 결합중 내부의 Arg-Val 결합이 끊어지면서 활성을 가지는 plasmin 으로 된다. 이 plasmin 이 fibrin clot 를 용해시키는 작용을 하게 된다.
이 plasminogen 을 활성화 시키는 활성인자는 체내의 여러 조직과 혈액내 에서 존재하는 활성인자와 bacteria 로부터 추출 할 수 있는 SK 와 staphylokinase, 뇨중에 존재하는 urokinase 등 여러 종류가 있다.
지금까지는 urokinase 에 의한 plasminogen 활성화에 대한 연구가 주로 진행 되었었다. 그러나 β-haemolytic bacteria 에서 추출 가능한 이 SK 는 작용 기작이 UK 와 현저히 다르다. 본 실험에서는 이 SK 가 촉매적 활성을 가지기 위해서는 human plasminogen 을 필요로 하는 allosteric 단백질로서 작용하는 SK 의 특성에 대해서 연구해 보았다. SK 의 량을 고정시키고 plasminogen 농도를 변화시켰을 때의 rate profile은 전형적인 saturation curve 를 나타내었다. 그러나 plasminogen 와 SK 의 량의 비가 작을 때는 rate profile 은 sigmoidity 를 나타내었다. 이러한 속도 반응론적인 결과와 다른 실험적 결과를 종합해 볼때 SK 와 plasminogen 은 빠른 속도로 결합하여 복합체를 형성하고 이것이 또 한 분자의 plasminogen 을 활성화 시키는 활성인자로 작용한다는 것을 알수 있었다. 즉 SK 그 자체는 활성을 가지지 않는 효소였으나 일단 plasminogen와 결합되면 활성을 가지게 되었다.
