The enzyme-linked immunosorbent assay using the so-called 'double-sandwich' technique was applied to determine $\underline{Clostridium}$ $\underline{botulinum}$type F toxin. By this technique, about 10 mouse intraperitoneal 50% lethal doses of type F toxin could be detected with negligible back-ground. This low back-ground reading was achieved with the use of phosphate-buffered saline containing 0.05% Tween 20 and 1% bovine serum albumin (PTB) as a diluent of bridging antibody and conjugate. For accurate determination of type F toxin in samples, addition of EDTA seems to be necessary. This assay permits no cross-reaction with botulinum toxins of other types tested.
$\underline{Clostridium}$ $\underline{botulinum}$ type F, strain Langeland 의 독소를 간단히 검출 할수 있는 면역학적 효소 방법을 연구하였다.
방법은 다음과 같다.
(1) Horse anti-type F serum을 polystyrene tubes 에 부착
(2) 각기 다른 양의 type F 독소가 들어있는 samples 가함.
(3) PTB로 희석시킨 rabbit anti-type F IgG를 가함.
(4) PTB로 희석시킨 sheep anti-rabbit IgG-HRP conjugate를 가함.
(5) HRP 에 대한기질 가함.
(6) 반응 정지시키고, A (449 nm) 기록
독소의 검출 한계는 10 mouse ip $LD_50$ 였으며, 이 방법을 다른 type의 clostridium strain에 적용 했을 때는 거의 반응이 안 일어났다. 또한 back-ground reading의 만족할 만한 결과는 PTB 의 사용시에만 얻어졌다.
