Urokinase, a proteolytic enzyme which is excreted from human urine, has the ability to convert plasminogen, the precursor of the fibrinolytic enzyme of blood plasma, into the active enzyme, plasmin. Urokinase showed a broad substrate specificity having hydrophobic amino acids, i.e., carbobenzoxy-L-tyrosine p-nitrophenylester, carbobenzoxy-L-phenylalanine p-nitrophenylester, and N-α-tosyl-arginine methylester.
The molecular weight of urokinase was estimated to be 41,000.
Both plasminogen activation and esterolytic activity of urokinase are thought to be attribution of the same active site from the fact that 6-aminohexanoic acid, which is an inhibitor of urokinase-plasminogen system, also inhibits the esterolytic activity of urokinase.
By the use of plasminogen-fibrinogen system, it was possible to detect very low content of urokinase.
사람의 뇨 중에 미량 존재하는 urokinase 는 혈장내의 전구물질인 plasminogen 을 plasmin 으로 활성화하여 fibrin 혈전을 용해시킬 수 있는 능력을 가진 단백질 분해 효소이다. 본 실험에서 p-nitrophenyl ester 가수분해에 의한 방법으로 urokinase 의 기질 특이성을 시험하여 urokinase 도 tyrosine과 phenylalanine 의 ester 에 대하여 가수분해 활성을 나타낸다는 사실을 알았다.
urokinase - plasminogen system 에 저해제로 알려진 6-aminohexanoic acid 가 urokinase 의 esterolytic activity도 역시 저해한다는 사실에서 plasminogen 활성화와 esterase 활성은 같은 활성부위에 의하여 나타난다고 볼수 있다.
그리고 plasminogen - fibrinogen system 를 이용하여 매우 적은 양의 urokinase 의 활성 측정이 가능함을 알았으며, urokinase는 일에 비교적 불안정한 효소임을 알았다.
Gel 여과 방법에 의하여 urokinase 의 분자량은 대략 41,000으로 추정된다.
