There are many types of brain diseases, such as an Alzheimer’s Disease (AD) and Parkinson’s Disease (PD). For the case of Alzheimer’s Disease, it is caused mainly by beta-amyloid accumulation. To treat such disease, anthocyanin can be used to protect beta-amyloid accumulation. In case for Parkinson’s Disease, it is caused by death of dopamine-generation cells in the midbrain region and many drugs are used for treatment. However, the possibility of the efficient drugs passing the BBB (Blood Brain Barrier) is low. Therefore, the use of drug carriers are an alternative method to treat brain disease. However, such treatment also requires sever-al standards - ability to transport molecules across the BBB, biocompatibility, proper circulation time, and right particle size and shape. Based on such reasons, Gold nanoparticles satisfy as one of the best drug carri-ers for treating brain diseases. Here, we report method to synthesize gold nanoparticles, where size can be controlled with asymmetric shape, resulting in sufficient use as a drug carrier for treatment of brain diseases.
To synthesize the gold nanoparticle, asymmetric gold nanoparticle’s seed was used, also known as polyol process. Gold (III) chloride trihydrate (HAuCl4o3H2O) was used for metal precursor and ethylene gly-col was used as a reducing agent, which donates electrons to gold ions (Au3+ + 3e- → Au0). Finally, for surfac-tant, also called the Capping agent, poly (diallyldimethyl ammonium chloride) (PDDA) was used which has the ability to attach {111} plane, explained by the Miller Index. By mixing N, N-dimethylformamide (DMF) and Gold (III) chloride trihydrate (HAuCl4o3H2O), followed by 5 hour sonication, gold nanoparticles were synthesized. To check the nanoparticles, morphology, UV-Vis absorption, zeta potential and applications through Ussing chamber were checked.
Through SEM (Scanning Electron Microscope) and TEM (Transmission Electron Microscopy), octa-hedral gold nanoparticles and asymmetric gold nanoparticles were successfully synthesized. Also, it was found that different experimental conditions, such as washing time, amount of gold ion’s solution, concentra-tion of DMF, and temperature, can affect the morphology of the gold nanoparticles. In addition through so-lution color observation, UV-Vis absorption, zeta potential and use of Ussing chamber, we confirmed the pos-itive use of asymmetric gold nanoparticle’s potential as use of nano drug carriers.
현재 알츠하이머 (치매), 파킨슨병 등 다양한 종류의 뇌 질환 질병이 알려져 있다. 치매의 경우, 베타-아밀로이드라는 특정한 단백질이 쌓여서 생긴다고 알려져 있는데, 치료의 한 방법으로 안토시아닌이라는 물질이 이 베타-아밀로이드의 축적을 방지해 준다고 알려졌다. 파킨슨병의 경우, 뇌의 가운데 부분에 도파민이라는 물질의 결핍이 원인으로 알려져 있다. 치료를 위해서, 현재 다양한 약물들이 쓰이고 있다. 하지만 뇌 안까지 이러한 약물들이 들어가는 효율은 낮은데, 이는 뇌 혈관 장벽 (BBB) 때문이다. 따라서, 뇌 질환 치료를 위한 나노 약물 전달체를 위해서는, 우선 뇌 혈관 장벽을 통과할 수 있어야 하고, 생체 적합해야 하며 적당한 크기와 모양을 가지고 몸 안에서 오랫동안 순환할 수 있어야 한다. 이러한 이유들을 바탕으로, 최근 금 나노입자가 나노 약물전달체로서 적합하다는 것이 알려졌다. 따라서 이 논문을 통해, 뇌 질환 치료용 크기 조절이 가능한, 비 대칭 금 나노입자 제조 기술을 소개하려 한다.
비 대칭 금 나노입자를 만들기 위해서 필요한 seed는, 폴리올 공정을 사용하였다. 우선, 금속 전구체로 Gold (III) chloride trihydrate (HAuCl4o3H2O)을, 환원제로는 에틸렌 글리콜 (Ethylene Glycol), 계면활성제로는 {111}면에 선택적으로 붙는 Poly (diallyldimethyl ammonium chloride) (PDDA)를 이용하였다. 이러한 seed를 N, N-dimethylformamide (DMF)와 적절한 양의Gold (III) chloride trihydrate (HAuCl4o3H2O)과 섞어서 5시간동안 자석을 이용해 섞어주면, 실험은 끝나게 된다. 결과를 확인하기 위해서, 실험 조건에 따른 표면의 변화와 더불어 색상, UV-Vis 흡광, 제타 포텐셜과 응용의 한 측면으로 우싱 챔버를 이용하였다.
주사전자현미경과 투과전자현미경을 이용해, 정팔각형 금 나노입자와 더불어 비 대칭 금 나노입자가 성공적으로 만들어졌음을 확인하였다. 또한, 다른 실험 환경, 예를 들면 다른 세척 횟수, 금 이온의 양, N, N-dimethylformamide (DMF)의 농도 그리고 실험 온도가 비 대칭 금 나노입자의 모양에 영향을 미친다는 점을 알아내었다. 그리고 용액의 색상, UV-Vis 흡광도, 제타 포텐셜 그리고 우싱챔버의 결과를 통해, 우리는 비 대칭 금 나노입자가 나노 약물 전달체로서 높은 잠재적 가능성을 가지고 있다는 결론을 내릴 수 있었다.
