Early diagnosis of cancer is of importance, considering that more than 90% of deaths in cancer pa-tients are attributed to metastasis. Current analytical methods for cancer including X-ray, MRI, CT, PET and ultrasound have limitations such as high-cost, prolonged time-consumption, and agonizing processes. Moreo-ver, these techniques can identify the cancer after its development. Therefore, the genetic based cancer diag-nostics has attracted attention due to its accuracy and reliability and is useful for cancer prognosis and diag-nosis. In this work, an advanced slidable microdevice for single nucleotide polymorphism (SNP) analysis has been developed to identify the cancer gene markers in the human genome. We targeted the mutation of tp53 gene which is located on the short arm of chromosome 17 and is highly related with cancer development. The slidable microdevice can perform the ligation-rolling circle amplification (L-RCA) reaction, and the fluores-cence detection of the RCA amplicons to identify the mutation. The chip consists of three glass layers: a channel wafer, a Ti/Pt electrode-patterned resistance temperature detector (RTD) wafer, and a slidable plate in which the reaction chamber was patterned. In the reaction chamber, the capture probe is chemically at-tached and the padlock probe is combined to the capture probe. If the target DNA is wild-type, the target is hybridized with the padlock probe and ligated. After ligation, the padlock probe becomes a circle template, and the RCA reaction proceeds. The amplicon is confirmed by fluorescence signal and reveals the SNP se-quence. In the case of mutation, the amplicon is not produced due to no ligation with the padlock probe. We simultaneously identify 5 mutations on a single microdevice with high efficiency.
대부분의 암환자들은 암의 전이현상 때문에 죽음을 맞게 된다. 따라서 암의 조기진단은 매우 중요하다. 현재 암을 진단하는 방법으로는 X-ray, MRI, CT, PET, 조직검사 등이 사용되고 있지만 이 방법들은 비용이 비싸고, 시간이 오래 걸리며, 복잡한 과정으로 환자에게 고통을 준다. 또한 이 방법들은 암이 발생하고 어느 정도 진행된 후가 되어야 진단이 가능하다는 단점을 가지고 있다. 이를 해결하기 위해 정확성과 신뢰성을 바탕으로 한 유전학 기반의 암 진단 방법이 주목을 받고 있다. 따라서 본 연구에서는 유전학 기반의 암 진단을 위해, 인간 유전자 암 마커를 유전자 다형성 분석을 통해서 확인해 보았다. 17번째 염색체 짧은 팔에 위치한 tp53유전자가 암의 발달과 큰 연관이 있기 때문에 tp53 의 변이 포인트 중 다섯 포인트를 목표로 잡았다. 모든 과정은 추가적인 초소형밸브나 초소형펌프가 필요 없는 이동형 마이크로디바이스 위에서 진행되었다. 이 발전된 디바이스는 챔버를 손으로 움직여 미는 간단한 컨셉을 통해 구축될 수 있었다. 이 마이크로디바이스는 마이크로채널이 패턴된 지름 4인치 유리웨이퍼 층, 온도를 조절하기 위한 Ti/Pt 전극 층, 반응이 일어나는 챔버 층으로 세 층의 구조를 하고 있다. 디바이스에서는 결찰 반응, 회전환 증폭 반응, 형광 신호 검출을 통하여 다섯 포인트의 변이 여부를 확인할 수 있었다. 만일 그 포인트가 변이상태라면, 결찰 반응이 일어나지 않기 때문에 회전환 증폭 반응이 일어날 수 없고, 형광 신호도 거의 나타나지 않을 것이다. 반대로 포인트가 정상상태라면 결찰 반응이 일어나고 회전환 증폭 반응이 뒤따르며, 그 결과 형광 신호를 얻을 수 있을 것이다. 모든 반응은 12개의 챔버에서 동시에 일어났으며 3시간안에 성공적으로 다섯 포인트의 변이 여부를 확인 할 수 있었다.
