Pharmacologically active components of ginseng, ginsenosides Compound K (C-K) and Protopanaxadiol (PPD) have been shown to have strong anticancer effects. However, their effect on glioblastoma has not been extensively evaluated. Further investigation on the mechanisms in which they induce cytotoxicity against tumor cell may be significant to develop them as alternative therapies from natural products.
Here, I investigated the anticancer effect and the molecular mechanisms of ginsenosides on glioblastoma. Among 18 ginsenosides tested, compound-K (C-K) and protopanaxadiol (PPD) reduced U251MG cell viability in dose- and time-dependent manner. This cytotoxic effect was accompanied by decreased expression of cell adhesion proteins including N-Cadherin and Integrin β1, which led to reduced phosphorylation of focal adhesion kinase. Furthermore, I observed the reduction of Cyclin D1 and subsequent G1-phase cell cycle arrest. I further demonstrated that C-K and PPD inhibited the phosphorylation of Akt and extracellular signal regulated kinase. Conversely, C-K and PPD stimulated phosphorylation of JNK and cleavage of PARP. These results collectively indicate that ginsenosides C-K and PPD may provide a new strategy to treat malignant glioblastoma that is quite resistant to conventional anti-cancer treatment.
인삼의 구성 요소 중 진세노사이드 Compound K (C-K)와 프로토파낙사디올계 (Protopanaxadiols, PPD type) 는 효과적인 항암작용이 있는 것으로 밝혀져 왔다. 그러나 이러한 진세노사이드가 뇌종양세포에서 어떠한 영향을 미치는지는 여전히 명확하게 밝혀지지 않고 있다. 진세노사이드가 종양세포에서 세포독성을 유발하는 작용기전을 밝히는 것은 천연약물을 이용한 치료방법 개발에 도움을 줄 것이다.
본 논문에서는 진세노사이드가 뇌종양에 미치는 항암효과와 분자생물학적인 작용기전을 밝혔다. 18개의 진세노사이드 중 Compound-K (C-K) 와 프로토파낙사디올계 (PPD)는 투여 용량과 시간에 따라 U251MG 세포 생존능력을 감소시킨 것을 확인할 수 있었다. 이러한 세포 독성 효과는 N-Cadherin 과 Integrin β1등의 세포 부착 단백질의 발현을 감소시켰으며 또한 FAK의 인산화반응을 감소시켰다. 더 나아가, 본 논문에서는 Cyclin D1의 감소와 그에 따른 G1기에서의 세포 주기 정지를 관찰하였다. C-K와 PPD는 Akt와 세포외 신호조절인산화효소의 인산화반응을 억제하였으나 JNK의 인산화반응과 PARP의 분해는 촉진시켰다. 이러한 결과들을 종합하여 볼 때, 진세노사이드 C-K와 PPD는 기존의 항암치료로 극복하기 힘든 악성뇌종양을 치료하는 데 새로운 방향을 제시할 수 있을 것이다.
