There is one factor-independent transcription terminator, called T7Tφ, in phage T7 genome for the phage-encoded RNA polymerase. Termination efficiency of this intrinsic terminator of 41 bp, surrounded by various DNA sequences, was determined in in vitro transcription reactions including linearized plasmid templates. Although the intrinsic terminator was always directly flanked by its upstream genomic sequence of 70 bp, its in vitro termination efficiency varied from 15% to 82%, while experimental error range is below 2%. The variation of the efficiency depended only on the entire sequence in a transcription unit, while the sequences outside of the unit did not have any effect. In all cases, however, the hairpin RNA structure of the terminator seems to be formed, as predicted by the energy minimization method of RNA folding. Also there seems to be no particular sequences that enhance or reduce the efficiency regardless of their positions.
Each termination efficiency did not vary upon the amount of transcripts produced, except in one case. Pulse labeling of transcripts in time course of in vitro transcription reaction revealed that the efficiency of the terminator in one particular construct (pGEM3ZT), but not in other constructs, was observed high (~60%) at an early time point but decreased exponentially until a low efficiency (~20%) persists.
Cotranscription of the pGEM3ZT with other templates resulted in reduced termination efficiency from the other templates. In order to identify the trans-acting element in the pGEM3ZT transcription reaction mixtures, after one-hour transcription the T7 promoter was destructed by Hinfl digestion, T7 RNA polymerase and Hinfl were removed by phenol extraction, and then the mixture was included in the second reaction with a second template and newly added polymerase. The resulting reduced termination efficiency of the second template strongly suggests that the trans-acting element is the RNA transcript from pGEM3ZT. Furthermore, trimming of the transcript from the 3' end indicated that even the 9-mer RNA of the sequence GGGCGAATT maintains the trans-acting antitermination activity. This sequence has no complementarity to any sequence of terminator-forming region. And the trans-acting antitermination activity had effects on the T7 terminator surrounded by any sequences. These results suggest that this transcript affects transcription ternary complex at terminator during termination process.
요소 독립적인 전사종결제의 종결효율은 종결제의 구조자체에 내재하는 것으로 알려져왔다. 우리는 파아지 T7 전사종결제를 전사촉진제를 가진 여러 플라스미드들에 클로닝한 후, 그 종결효율을 측정하였다. 그러나, 놀랍게도 종결효율은 기존에 알려진 약 80%와는 큰 차이가 있었다. 이에 우리는 종결효율에 미치는 요인들을 조사 하였다. 이 종결효율은 전사단위를 벗어난 주변 DNA 염기서열 및 전사촉진제의 염기서열에 영향을 받지 않았으며, 전사단위 내의 RNA 머리핀 구조에도 영향을 받지 않았다. 이어 전사개시 부분에서 전사종결제 앞까지 각 부분의 염기서열을 바꾸며 종결효율은 조사하였다.
그 결과, 종결효율은 전사단위 내의 전체 염기서열에 따라 15%에서 82%까지 크게 변화하며, 종결효율을 높이거나 낮추는 특정한 염기서열은 발견되지 않았다.
또한, 다른 플라스미드와는 달리 pGEM3ZT의 종결효율이 만들어진 RNA의 양에 따라 달라진다는 것을 발견하였다. 펄스 표지로 반응시간에 따른 종결효율을 조사한 결과, pGEM3ZT는 반응 초기에는 높은 종결 효율을 가지나(약 60%), 시간이 지남에 따라 매우 급격하게 감소한 후 일정한 종결효율(약 20%)을 보이는 것을 발견하였다.
pGEM3ZT의 전사반응의 생성혼합물이 그 자체의 전사종결 효율을 낮출 뿐만 아니라, 다른 플라스미드의 종결효율도 낮추는 성질을 가지고 있는 것이 발견되었다. 이에 pGEM3ZT의 전사생성 혼합물을 분석하여 pGEM3ZT의 전사물이 트랜스로 작용하는 반-종결 성질을 가지고 있으며, 전사염기서열 중에서도 전사개시부분의 GGGCGAATT가 이 성질을 가지고 있음이 발견되었다. 이 트랜스로 작용하는 반-종결성질은 이 연구에서 사용된 여러 플라스미드들 중에서는 pGEM3ZT에서와 그 유도체에서만 발견되었으며, T7 전사종결제를 가진 시험해본 모든 플라스미드들의 종결효율을 감소시켰다. 이로부터 우리는 pGEM3ZT의 GGGCGAATT의 염기서열을 가지는 전사물이 전사종결 단계에서 종결효율에 영향을 미친다는 것을 알 수 있었다.
이 연구로부터 우리는 전사종결제 자체의 구조가 전사종결 효율을 결정하는 유일한 요소가 아니라는 사실을 밝힐 수 있었으며, 또한 생성된 전사물에 의한 반-종결 현상이 새롭게 발견되었다.
