To develop enzymatic methods of glycosidic-bonds formation, glycosidases in organic solvents were utilized as a catalyst of condensation betweem sugars, and sugar and alcohol. In these system, organoboronic acids, for example, phenylboric acids and butylboric acid were introduced to solubilize sugars in organic solvents. Organoboronic acids in these system were condensated with sugars to give sugar-boronic esters and condensing water. The condensation in microaqueous two-phase system was critically influenced by the amount of water and organoboronic acids. It has been investigated the effects of water and organoboronic acids on the formations of disaccharides and alkyl-β-D-glycosides by the quantitative HPLC analysis and Karl-Fisher titration of water contents in organic phase. Various organic solvents having different water solubilities were examined to find optimal reaction medium in the syntheses of ethyl-β-D-glucoside and ethyl-β-D-galactoside. Also, optimal water contents in aqueous phase were evaluted in terms of the Q value defined as following : mole of water divided by mole of sugar in aqueous phase. Q values of optimal condensation increased on the increase of solubility of solvent in water. Q values of optimal condensation were different according to alcohols i.e., in case of (1) water miscible alcohol : Q value slightly increased on the increase of nucleophilicity, and (2) water immiscible alcohol : Q value increased greatly on the increase of hydrophobicity. As a whole, Q values were dependent on the activity of water and alcohols.
The concentration of products and reactants concentration in organic phases were affected by the amounts organoboronic acid and water in the reaction system. In the presence of phenylboronic acid, glucose was more soluble in organic solvents than alkyl-β-D-glucoside, whereas less soluble galactose than alkyl-β-D-galactoside. Organoboronic acids in the formation of ethyl-β-D-galactoside increased more than five folds of product concentration in organic solvents. Primary and secondary alcohol as nucleophiles to the enzymatic reaction with monosaccharides give their alkyl-β-D-glycoside in these reaction system, but no tertiary alkyl-β-D-glycosides were formed. All alkyl-β-D-glycosides from enzymatic syntheses were optically pure β-anomers, which were confirmed by $^{13}C$-NMR analysis. Finally, n-pentenyl-glyosides, as a glycosyl donor molecules in chemical synthesis of glycosides, were synthesized through these methods.
글리코시딕 결합의 효소적인 합성 방법을 개발하기 위하여, 유기용매중의 글리코시다제를 당과 알콜간의 축합을 위한 촉매로 선택하였다. 이 반응계에 페닐붕산이나 부틸붕산과 같은 유기붕산을 도입하여 당를 유기용매중으로 녹일수 있었다. 이 반응계에서 유기붕산은 당들과 축합반응하여 당유기붕산 에스테르와 축합수를 형성한다. 이때 축합수에 의해 형성된 미수계에서의 축합반응은 물과 유기붕산의 함량에 의해 크게 영향을 받았다. 이당류와 알킬글리코시드의 형성에 미치는 이 영향을 고속 액체크로마토그라피에 의한 생성물 분석과 칼휘셔법에 의한 수분함량 분석을 통하여 조사하였다. 모델 화합물로서 베타형의 에틸글리코사이드와 에틸갈락토사이드의 형성에 영향을 주는 요인들을 조사하였다. 최적의 반응용매을 조사하기 위해 각각 물의 용해도가 다른 여러가지 용매들을 조사하였다. 물층에 있어서 물함량을 당함량으로 나눈 값을 Q 값으로 정의하고 이 값으로 최적의 물함량을 조사하였다. 물에 대한 유기용매의 용해도가 증가할수록 최적의 Q 값은 증가 하였다. 최적의 Q 값은 알콜에 따라 달라졌는데 물과 섞이는 알콜일때는 친핵성이 증가할수록 증가하였고, 물과 섞이지 않는 알콜은 소수성이 증가할 수록 Q 값이 증가하였다. 전체적으로 최적의 Q 값은 물과 알콜의 활동도에 따라 변하였다. 본 반응계에 유기붕산을 도입하여 유기층의 탄수화물 농도를 증가시켜, 추출 촉매 반응을 시도하였다. 유기층의 반응물과 생성물의 농도는 페닐붕산과 물함량에 따라서 변하였다. 유기붕산 존재 하에서 포도당은 알킬화 포도당보다 유기용매에 잘 녹았고, 유당의 경우는 반대였다. 그러므로 에틸유당의 합성시, 유기붕산을 넣어 줌으로서 유기층의 에틸유당의 농도를 5 배 증가시킬수 있었다. 메탄올부터 펜탄올까지 여러가지 알콜들의 반응성을 조사한 결과, 1 차 2 차 알콜들이 효소의 기질로 사용될 수 있었으며 3 차 알콜은 작용하지 못하였다. 합성된 모든 알킬글리코사이드들은 탄소핵자기공명 분석 결과 순수한 베타형으로 확인되었다. 본 연구의 방법으로, 최근에 개발된 탄수화물 합성에 중요한 시발물질이며, 화학적 방법으로 여러 단계를 거쳐 합성되는 n-펜텐닐글리코시드들를 포도당과 유당으로부터 각각 100 mM과 50 mM의 농도로 직접 합성하였다.
