The mortality rate by coronary artery disease has been increased. Atherosclerosis is a disease caused by thickened artery wall as a result of the accumulation of calcium and fatty materials. Acute coronary syndrome, a type of myocardial infraction, is a disease that is caused by rupture or erosion of atherosclerotic plaque. These vulnerable plaque is characterized as thin fibrous coat and lipid below the coat, therefore, lipid detection is important to diagnosis of the vulnerable plaque to prevent coronary artery disease.
Optical frequency domain imaging (OFDI) is one of the biomedical imaging techniques that is composed of a light interferometer and a wavelength swept laser. High sensitivity of OFDI system enables high speed imaging of human arteries. Because of high speed imaging and high axial resolution, OFDI system provides specific structures of human arteries. However, OFDI system only provides the intensity information that is proportional to the amount of scattered light from the tissue, therefore, it is necessary for experts to classify tissue.
In this thesis, we noted that each composition of atherosclerotic plaque has different characteristics such as scattering and absorption, in accordance with the wavelength of the light. Especially, lipid has strongly attenuated in the wavelength range of 1200 nm. We demonstrated dual wavelength-band spectroscopic OFDI system in the wavelength ranges of 1191 nm ± 42.5 nm and 1292 nm ± 49 nm and calculated the attenuation coefficient of the sample. We selected mayonnaise as a lipid phantom and made a scattering phantom for comparison. We performed imaging of mayonnaise and scattering phantom and analyzed the intensity profile. We compensated effects of coherence and objective lens and eliminated the scattering effect. We calculated the attenuation coefficient by liner regression and differentiation and represented attenuation coefficients as an image. As a result, we proved the possibility of distinction of lipid between dual wavelength bands.
관상성 동맥 질환에 의한 사망률은 점점 증가하고 있다. 아테롬성 동맥 경화증은 칼슘이나 지방 성분의 침착에 의해 혈관 벽이 두꺼워짐으로써 발생하는 질병이다. 급성 관동맥 증후군은 심근 경색의 한 종류로, 동맥 경화반의 파열이나 미란에 의해 발생한다. 이러한 취약성 경화반은 얇은 섬유막과 섬유막 하부의 지질 성분을 특징으로 하는데 따라서 지질 성분의 검출은 관상성 동맥 질환을 방지하기 위한 취약성 경화반 진단에 있어 중요하다.
광 주파수 영역 이미징(OFDI)는 생물의학 이미징 기술 중 하나로 간섭계와 파장 변환 레이저로 구성된다. OFDI 시스템의 높은 민감도는 사람 혈관을 빠른 속도로 이미징이 가능하게 해준다. 이러한 빠른 속도의 이미징과 높은 축 방향 해상도 때문에 OFDI 시스템은 혈관의 특정한 구조를 제공한다. 그러나 OFDI 시스템은 오직 조직에서 산란되어 온 빛의 양에 비례하는 빛의 세기 정보만을 주기 때문에 조직을 구분하기 위해서는 전문가가 필요하다.
본 논문에서, 우리는 동맥 경화반의 각 성분이 빛의 파장에 따라 산란이나 흡수와 같은 서로 다른 특징을 가진다는 점에 주목하였다. 특히, 지질은 1200 nm 파장 영역에서 강한 감쇠가 발생한다. 우리는 1191 nm ± 42.5 nm and 1292 nm ± 49 nm 파장 영역을 가지는 이중 파장 분광학적 OFDI 시스템을 제작하였고, 샘플의 감쇠 계수를 계산하였다. 마요네즈와 산란 팬텀을 이미징하고 그것들의 세기 정보 프로필을 분석하였다. 우리는 가간섭성과 대물 렌즈에 의한 효과를 보정하였고 산란에 의한 효과를 제거하였다. 그리고 선형 회귀법과 미분법을 사용하여 감쇠 계수를 계산하고 이렇게 계산된 감쇠계수를 이미지로 표현하였다. 결과적으로 우리는 이중 파장 사이에서 지질 성분의 구분 가능성을 확인하였다.
