Traditional Chinese medicines are regarded as promising source for drug development, since they are relatively safe and their therapeutic effect are previously known, thanks to accumulated trial experiences for over-thousand years. However, its integrated mechanism of action in multi-component level is still unknown.
In this research, we developed a system that predict the effect of the multicomponent drug based on multi-target analysis of molecular information. We have integrated biological databases (KEGG, ChEMBL) to obtain drug-related pathways. Next, pathway entities were divided into more specifically defined smaller sub-pathway named product oriented sub-pathway (PSP). From PSP-linked to metabolite sets, drug-affected terms in physiological level were obtained through co-occurrence analysis in PubMed.
We have performed a case study for the complex herbal drugs composed of Corydalis tuber and Pharbitis seed. Resulted physiological function terms showed significant similarity with previously known functions of the TCM ingredients.
천연물신약은 그 개괄적 효능과 생물독성 여부가 오랜 기간 축적된 경험을 통해 검증되어있으므로, 개발 과정에서 상대적 장점을 가져 신약개발 분야에서 각광받고 있다. 반면 다성분 천연복합물을 그대로 활용하는 특성상 효능성분 및 분자 단위 기작을 특정하기 난해하고, 약제 성분의 표준화를 이루기 어렵다는 단점을 보인다. 본 연구는 생물학적 경로 데이터베이스를 이용한 다중표적 분석 시스템을 통하여 다성분 천연물 신약의 효능을 예측하는 것을 목적으로 하였다.
본 연구에서는 다성분 복합물로 이루어진 천연물신약의 효능을 예측하기 위해 생물정보데이터베이스 간의 연동, 경로 네트워크 상에서 결과 대사물을 기준으로 편성된 ‘결과대사물 기반 세부경로’(PSP)의 추출, 연관점수에 따른 결과 대사물 간의 정렬, 텍스트 마이닝에 기반한 상위 수준 효능 유추 등의 요소를 적용하였다. 또한 이와 같이 구축된 모델에 현호색과 견우자의 복합물로 이루어진 천연물신약의 정보를 적용하여 봄으로써, 그 예측결과가 알려진 약재의 효능과 합치함을 확인하였다.
