Biodiesel is one sustainable alternative to fossil-based liquid fuels. Among various feedstock, oleaginous microbes, cells able to accumulate lipid over 20% of their dry masses, have received increasing attention due to their exceptionally high lipid productivity. Microalgae and heterotrophic yeasts are two of the most promising and thus studied cell types. Microalgae generally grow photoautotrophically and hence accumulate lipid through photoautotrophic metabolism, only requiring carbon dioxide as a sole carbon source and light as an energy source. Slow growth and low final cell density are two fatal disadvantages of these photoautotrophs in relation to their usage as fuel feedstock. Oleaginous yeasts, on the other hand, have much higher growth rate and lipid productivity. The final cell density can even reach a commercially viable level relatively easily. One critical caveat is their requirement of growth substrates, which constitutes approximately half of total fuel production cost. Accordingly, one success key of heterotroph-based fuel production is to find sufficiently cheap feedstock. It is equally important to obtain potent yeast trains. These two critical issues were therefore be addressed in this master thesis.
First, several oleaginous yeasts were isolated particularly from carbon-rich environments such as mine drainage, spoil, and soil, based on my assumption that in this condition oil-degrading microbes proliferate. My working hypothesis for this was that good oil degraders are also able to accumulate lipid well, as their lipid synthesis must be metabolically linked to lipid degradation. After isolation, each of the 4 isolated yeasts was sequenced for identification. Among these 4 kinds of yeasts were selected for detail characterization. Growth rate was employed as parameter, as it, together with lipid content, would be one determining factor for commercialization. Various culture conditions, such as carbon source, temperature, and pH, were examined. Lipid content was also quantified with additional variables of C/N ratio and salinity. Consequently, two isolates were found to be oleaginous and named as Candida pseudolambica KCTC 27547 (Max. lipid content: 45.0% of dry mass) and Issatchenkia occidentalis KCTC 27551 (Max. lipid content: 23.0% of dry mass).
In addition to the isolation of our own promising strains, low cost feedstock was also sought. One potential feedstock is livestock wastewater (LW), as it is cheap, carbon- and nutrients-rich, and abundantly available. To disintegrate organics and nutrients existing in less utilizable, complex forms and at the same time inactivate indigenous microbes, raw LW was pretreated by chemical and/or physical means. Phosphor, which is a precious element contained in the wastewater at a high amount (up to 460 mg/L), was recover as struvite (NH4MgPO4o6H2O) by supplying Mg2+ and increasing pH (higher than 9) by means of the addition of MgO. As physical means, vigorous shaking, ultrasonication, and hydrodynamic cavitation (HC) were adopted and compared for in terms of phosphorus removal and disintegration degree (DD). It was found that with a physical treatment the formation of phosphate struvite was synergistically accelerated pH between 9 and 11. HC caused 100% phosphorus removal, and 80% of DD value. This powerful physical method also rendered ammonia significantly stripped out from the liquid phase. The gaseous ammonia can be easily captured and used for other useful purposes. Finally, the supernatant of HC-based pretreated LW was utilized as a growth medium for the cultivation of the isolated oleaginous yeasts. Tested media were prepared as follows: (1) one diluted with distilled water (DW) (pretreated LW/DW-no dilution, 1/1, 1/2, 1/4, and 1/8), (2) one supplemented trace metals, (3) one supplemented with vitamins, (4) one supplemented with both of trace metals and vitamins, and (5) one diluted with defined medium without any nitrogen source (DM-n). Each medium was also supplemented with a carbon source to regulate C/N ratio (C/N 20/1, 50/1, 75/1, and 120/1). Through these growth experiments, the possibility of use of pretreated LW could be verified. In medium amended with trace metals and vitamins oleaginous yeasts grew best. DM-n was found to support the highest lipid: Maximum lipid content of 35.4% of dry mass from Candida pseudolambica KCTC 27547 and 28.9% from Issatchenkia occidentalis KCTC 27551.
급속한 산업발달과 인구 증가로 에너지 수요가 커지고 있지만 에너지생산에 큰 비중을 차지 하는 화석연료의 매장량에는 한계가 있어 새로운 대체 에너지 자원이 필요하다. 뿐만 아니라 화석연료의 무분별한 사용으로 이산화탄소, 질소산화물, 황산화물 등 환경오염도 가속화되고 있는 실정이다. 따라서 환경오염을 최소화하는 천연 대체 에너지 원에 대한 필요성이 대두되고 있다. 그 중, 바이오 에너지는 바이오 매스를 기반으로 한 대체 에너지로써 바이오 에탄올, 바이오 디젤, 메탄, 수소연료 등을 포함한다. 바이오 매스는 자원의 양이 무한하다는 장점을 가지고 있으며, 생산 에너지 밀도가 높아 산업에 실용화가 쉽다는 장점을 가지고 있다.
바이오 매스의 종류는 크게 식용작물을 기반으로 한 1세대, 비 식용 작물인 목질계 바이오 매스를 기반으로 한 2세대, 미생물을 기반으로 한 3세대로 분류된다. 그러나 곡식, 농작물을 기반으로 하는 1세대 바이오 매스의 경우 식량자원의 부족과 가격 인상을 초래할 수 있다는 문제점을 가지고 있으며, 2세대 바이오 매스의 경우 과도한 산림자원의 파괴를 초래할 수 있다는 단점을 가지고 있다. 따라서 3세대 바이오 매스가 유망한 차세대 에너지 원료로 주목 받고 있다. 3세대 바이오 매스의 종류에는 미세조류, 효모, 박테리아, 곰팡이 등이 있으며, 미생물 내의 축적된 지질을 추출하여 연료로 사용 가능하다. 미생물 내의 지질은 산/염기 촉매를 바탕으로 알코올과 에스테르화 반응을 일으켜 Fatty acid methyl ester (FAME)을 형성하는데, 이는 화석연료인 디젤과 비슷한 조성을 가지고 있어 산업에 바로 이용 가능하다는 장점을 가지고 있다. 미세조류와 효모의 경우 미생물 자체의 크기가 크고 지질 함유량이 높다. 미세조류의 경우 추가적인 영양분 없이 광합성을 통해 생장 및 지질축적이 가능하다는 강점을 가지고 있으나 지질 함유 효모의 경우 적절한 영양분이 공급되면 미세조류에 비해 오염이 적어 생산이 용이하고 고농도 배양이 가능하여 대량생산이 용이하다. 따라서 지질 함유 효모의 최적 성장조건을 찾아 생산량을 극대화 시키고 저가 기질을 통해 지질 함유량을 극대화 시키는 공정이 필요하다.
본 연구에서는 바이오 디젤의 생산을 위해 지질 함유 효모를 분리하고, 성장 및 지질 축적에 적합한 배양 조건을 찾는 것을 목표로 하였다. 다량의 지질을 함유하는 미생물을 찾기 위해 탄소영양분이 풍부한 석탄 광산의 토양 및 배수에서 샘플을 채취하였으며, 샘플 내에 존재하는 지질 함유 미생물 만을 분리하기 위해 디젤만을 기질로 주어 생존 가능한 미생물을 선택적으로 분리하였다. 이는 디젤을 생장에 이용할 수 있는 미생물의 경우 생장 메커니즘 내에서 디젤을 이용가능하고, 동시에 축적도 가능하다고 알려져 있다. 분리된 미생물을 기반으로 하여 바이오 디젤 생산의 산업화를 위한 대량생산의 타당성을 고려하여 성장속도 및 지질 함유량을 측정하였다. 성장속도 및 지질함량을 결정하는 요인으로는 배지 탄소원 및 염의 종류 및 농도, 미생물 성장 온도, pH등을 고려하였다. 이를 통해 각 분리된 미생물들의 지질 함량을 극대화 하고 기존의 바이오 디젤 원료로 주목 받던 식물성 기름 함유량과 비교하여 바이오 디젤 생산의 원료로써의 가능성을 알아보고자 하였다.
또한, 분리한 지질 함유 효모를 기르기 적합한 저가 기질로써 축산폐수를 선정하고 전처리를 통해 부산물을 증가시키고 미생물의 기질로써 효율을 증가시키는 연구를 진행하였다. 축산폐수는 화학적 처리를 통해 방류되거나 톱밥과 혼합하여 퇴비로 사용되고 있으나, 비용이 많이 들 뿐만 아니라 독성 등에 대한 한계를 가지고 있다. 따라서 폐기물을 재활용 하는 점에서 친환경적이며 저가로 대량을 확보할 수 있다는 장점을 가지고 있다. 또한, 전처리 과정에서 발생하는 암모니아 가스와 인-결석은 비료, 건축물의 자재, 에너지원으로 사용 가능하여 경제적 이점도 가지고 있다. 축산폐수 내의 독성을 가지는 인 성분을 제거하기 위해 수용액 상에서 염기를 띄는 MgO를 첨가하여 인-결석 형성을 유도하는 Mg2+를 보충해주는 동시에 결석 형성에 적합한 pH조성해 주었다. 그 후, 혼합, 초음파, 공동화 현상을 이용하여 물리적 전처리를 진행하여 polymer와 cell을 깨트려 미생물이 이용가능 한 성분을 극대화 시켰다. 인 제거율과 Disintegration degree(DD) 값을 측정하여 전처리 효율을 비교하였고 질소 성분의 변화를 측정하여 전처리 과정에서의 암모니아 가스 발생량을 추정하였다. 가장 높은 효율을 보이는 공동화 현상을 이용한 전처리 후 상등액 만을 미생물의 저가기질로 사용하였다. 축산폐수 자체를 배지로 사용하거나 trace metal과 비타민 성분들을 추가하여 미생물의 성장에 적합한지 확인하였다. 또한 전처리 후 높은 질소함량을 보이는 축산폐수를 배지의 질소원으로 사용하여 성장속도를 측정하고 지질 함량을 비교하여 미생물의 저가 기질로써의 가능성을 확인하였다.
