This thesis presents a frequency-dependent cell impedance analysis chip for use in cancer and normal cell discrimination. The previous cell impedance analysis chips for flowing cells do not allow enough time for cell-to-electrode contact to monitor frequency-dependent impedance response. Another type of the previous cell impedance analysis chips for the cells clamped by membranes need complex sample control for making stable cell-to-electrode contact. We present a new impedance analysis chip using the SU8 microchamber array, on which a PDMS cover is placed to make stable cell-to-electrode contact for the individual cell trapped in each microchamber; thus achieving frequency-dependent single-cell impedance analysis without complex sample control.
We design and fabricate the array of 24 microchambers, each having size of 32x32x12 μm3, so that the PDMS cover traps individual cells in the microchambers to make the cell-to-electrode contact diameter as 80% of 20μm-diameter cells. Using the fabricated chip, we have measured and compared the impedance (magnitude and phase) response of human lung cancer cells (H358, A549) with the response of normal lung cells (the human brachial/tracheal epithelial cells of NHBE) in the frequency range of 200Hz to 2MHz for the AC signal of 100mVp-p. Compared to the normal cells, the cancer cells show impedance response difference: the magnitude difference of 28.30kΩ~151.67kΩ in the frequency range of 95.6 kHz~2MHz and the phase difference of 1.97°~16.83° in frequency range of 4.37 kHz~2MHz, respectively. In particular, the normal cells shows significant phase difference of 14.24°~16.83° comparing to the both cancer cells in the frequency range of 15.0 kHz~27.8 kHz.
It is demonstrated experimentally that the impedance analysis chip performs frequency-dependent cell impedance analysis by making stable cell-to-electrode contact with simple sample control; thereby applicable to the normal cell and cancer cell discrimination for further advanced cancer diagnosis and prognosis research.
본 논문에서는 주파수에 따른 단일세포의 임피던스 특성 분석칩을 제안하고 암세포와 정상세포의 구별에의 적용을 보였다. 세포 분석 기술 중 병이있는 세포와 정상세포를 구별하는 것은 하나의 큰 이슈이며, 화학적인 라벨링이 필요하지 않고 비침습적인 방법으로 살아있는 세포를 분석 가능하기 때문에 임피던스 분석은 세포 분석에 널리 이용된다. 기존의 임피던스 분석칩은 세포가 전극에 머무는 시간이 짧아 주파수 스윕분석이 어렵거나, 세포를 전극에 위치시키기 위해 복잡한 시료의 제어가 필요한 단점이 있다. 반면, 본 연구에서 제안하는 방법은 격벽을 갖는 챔버에 고정판을 덮어 단일 세포를 전극에 위치시킴으로써 안정적인 접촉이 가능하며, 따라서 복잡한 시료의 제어 없이 주파수에 따른 임피던스 특성 분석이 가능하다.
제안된 임피던스 특성 분석칩은 단일세포가 위치할 수 있는 24개의 챔버와 세포를 전극에 고정하는 PDMS 고정판으로 구성되며, 챔버당 직경 20um 내외의 단일 세포가 위치할 수 있게 챔버의 폭과 길이는 각각 32um로 설계하고 높이는 세포가 전극에 닿는 부분의 직경이 세포 크기의 80%가 되도록12um로 설계하였다.
본 실험에서는 정상세포(NHBE)와 폐암세포 두 종(H358, A549)을 이용하여 세포의 임피던스 특성분석 및 그에 따른 정상세포와 암세포의 구별을 실험적으로 검증하였다. 정상세포와 폐암세포는 임피던스의 크기로 95.6kHz~2MHz 범위에서 28.30kΩ~151.67kΩ 의 차이를 통해 구별이 가능하며, 임피던스의 위상으로는 4.37kHz~2MHz의 범위에서 1.97°~16.83° 의 차이를 통해 구별이 가능하였다. 특히, 정상세포는15.0kHz~27.8kHz 영역에서 2종의 폐암세포 모두와 14.24°~16.83° 의 차이가 나는 정상세포 고유의 위상차를 갖는 것을 확인하였다. 또한 세포의 등가모델을 바탕으로, 정상세포의 임피던스는 2.71xΩ, 정전용량은 7.01nF, 폐암세포 2종(A549, H358)의 임피던스와 정전용량은 각각 2.55xΩ, 2.95nF, 2.26xΩ, 3.30nF으로 측정 가능하였다. 따라서, 격벽을 갖는 챔버에 고정판을 덮는 방법으로 단일세포를 전극에 위치시켜, 복잡한 시료의 제어 없이 주파수에 따른 임피던스 특성 분석과, 그에 따른 암세포와 정상세포의 구별이 가능함을 실험적으로 검증하였다.
본 연구에서는 격벽을 가진 챔버와 PDMS 커버를 이용해 세포-전극간 안정적인 접촉을 구현하여 간단한 샘플 취급으로 주파수에 따른 임피던스 분석이 가능한 분석칩을 설계, 제작하고 그에 따라 암세포와 정상세포의 구별이 가능함을 검증하였다. 따라서 제안된 분석칩은 암세포와 정상세포의 구별과 나아가 세포의 임피던스 특성 관찰 및 암 진단에 활용할 수 있다.
