Functional polymer thin films is applied to much of modern science and technology community. Our group can easily fabricate functional polymer films via initiated chemical vapor deposition (iCVD) method.
One study is related to “a vapor deposited thermo-responsive polymer film, poly(N-vinylcaprolactam) (pNVCL) for cell sheet engineering”. Poly(N-vinylcaprolactam), pNVCL, is a thermo-responsive polymer. pNVCL is known to be nontoxic, water soluble, and biocompatible. Here, pNVCL was synthesized successfully by use of a vapor-phase process, initiated chemical vapor deposition (iCVD). Fourier transform infrared spectroscopy (FTIR) results showed that a radical polymerization took place from N-vinylcaprolactam (NVCL) monomer, without damaging the functional caprolactam ring. A sharp lower critical solution temperature (LCST) transition was observed at 31 oC from the iCVD pNVCL film. Thermo-responsive intelligent surfaces exhibited a hydrophilic/hydrophobic alteration with external temperature change, which, enabled a thermally modulated attachment and detachment with cells. The advantage of this system is that cells cultured on such thermo-responsive surfaces could be recovered as a cell sheet, simply by lowering the temperature without conventional enzymatic treatment.
Another study is related to “peptide immobilized iCVD pGMA surface as a cell culture substrate and control of hMSC’s differentiaion into osteogenic lineages”. Cell-substrate interactions are crucial for controlling diverse cellular behaviors. In particular, surface topographical effects of culture substrate are known to modulate adhesion, alignment, proliferation, and differentiation of stem cells. In this study, we investigated the effects of nanoscale topographies on the behaviors of human mesenchymal stem cells (hMSCs). Human adipose tissue-derived MSCs were cultured on various shapes of polyurethane acrylate (PUA) nanopatterned surfaces grafted with poly(glycidyl methacrylate) (pGMA) by initiated chemical vapor deposition (iCVD) technique. The surfaces of PUA substrates were modified with epoxy group of pGMA for potential grafting of bioactive molecules. Given that pGMA contains epoxy group that can readily bind to amine functional group, diverse bioactive molecules including growth factors and adhesion peptides can be easily immobilized onto the pGMA-PUA nanopatterned surfaces. Therefore, combinatorial effects of surface topography and bioactive molecules will be examined for enhancing proliferation and lineage-specific differentiation of hMSCs in this study.
기능성 고분자 박막은 과학과 기술 분야에서 아주 널리 응용되는 물질로 활발히 연구되고 있다. 우리 연구실에서는 개시제를 이용한 화학기상증착 방법을 통해 기능성 고분자 박막을 쉽게 제작할 수 있는데, 이러한 기능성 고분자를 이용하여 바이오 메디컬 분야에 응용하는 두 가지 연구에 대해 소개하도록 하겠다.
PNVCL (Poly(N-vinylcaprolactam))은 무독성, 가용성, 온도감응성, 생체적합성 등의 다양한 성질을 가지는 고분자이다. 온도감응성 고분자인 pNVCL은 개시제를 이용한 화학 기상 증착 방법을 통해 본 연구실에서 성공적으로 첫 합성을 해내었고, 이렇게 합성된 고분자는 알려진 그대로 31-34 oC 범위의 저임계 용액 온도를 가진다. Fourier 변환적외분광법을 이용해서 pNVCL의 caprolactam 고리에 손상을 입지 않고, 탄소-탄소 이중결합 부분이 열리면서 중합이 이루어졌다는 것을 증명하였다. 고분자의 특성은 Fourier 변환적외분광법, X-선 광전자 분광법, 겔투과크로마토그래피, 주사전자현미경, 자외선-가시광선 분광법, 물접촉각 등의 다양한 방법을 통해 측정하였다. 우리가 합성해낸 pNVCL 온도감응성 지능형 표면은 외부의 온도 변화에 따라 친수성/소수성 변화를 보였고, 이러한 성질을 이용하여 세포의 탈부착을 가능하게 할 수 있었다. 세포 실험을 통해 세포의 탈착에서, 전통적인 방법의 효소를 이용하지 않고 pNVCL 기판에서 세포를 키운 다음 단지 온도를 낮추는 것만으로도 세포를 떼어낼 수 있다는 결론을 얻었다. 여기서 한 가지 중요한 점은, 효소를 이용하면 세포가 각각 개별적으로 떨어지게 되는데, 온도감응성 기판을 이용하면 세포층을 그대로 유지하며 하나의 판으로 떼어낼 수 있다는 것이다. 이렇게 세포판으로 떼어낸다는 것은 세포판 응용공학에 이용할 수 있어 굉장히 유의미한 일이다.
세포-기판 상호작용은 다양한 세포의 거동을 조절하는데 매우 중요하다. 특히 세포배양기판의 표면 형태는 줄기세포의 부착, 정렬, 증식, 분화 등을 조절한다고 알려져 있다. 이 연구에서, 우리는 사람의 중간엽줄기세포가 나노 스케일의 표면형태에 따라 어떤 영향을 받는지 조사해 보기 위해, 다양한 형태의 polyurethane acrylate (PUA) 나노-패턴을 결함없이 만들어 내었다. 그 위에 개시제를 이용한 화학기상증착 기술로 poly(glycidyl methacrylate) (pGMA)를 균일하게 덮어 주었고, 에폭시기를 포함하는 pGMA는 아민 작용기와 쉽게 결합할 수 있는 성질을 지니고 있기 때문에 성장인자와 부착펩타이드들을 포함하는 다양한 생리활성 분자들이 pGMA-PUA 나노패턴된 기판에 쉽게 고정화될 수 있었다. 이러한 방식을 통해 제작된 다양한 형태의 BMP2-pGMA-PUA 나노패턴 기판 위에 사람의 지방조직으로부터 유래된 중간엽줄기세포를 배양하였다. 이 연구를 통해 표면형태와 생리활성 분자 두 가지 변수의 결합에 의한 시너지 효과가 인간의 중간엽줄기세포의 특정 계통으로의 분화와 증식에 어떤 영향을 미치는지를 조사해 보았는데, 우리는 BMP-2 펩타이드와 나노패턴 형태의 결합에 의해 골형성 유도인자 없이도 중간엽줄기세포가 뼈발생 세포로의 분화를 유도한다는 것을 확인할 수 있었다.
이를 통해 우리가 합성한 기능성 고분자 박막을 세포에 독성 없이 적용할 수 있다는 것을 확인하였고, 다양한 작용기를 가지는 고분자를 합성하여 바이오 분야에서 여러 방면으로 유의미한 결과를 얻을 수 있음을 알 수 있었다.
