Electrical signals are the fundamental information carriers in the nervous system. To investigate neural networks, neuroelectronic interfaces are relevant. Metal microelectrodes have been broadly used for recording and stimulation of nervous system. The performance of microelectrodes are determined by low tissue-electrode interfacial impedance and high charge injection limits. Nanoscale surface engineering has been studied to improve the properties of microelectrodes. In this study, a novel microelectrode have been developed which has unique gold nanograin structures on its surface. The developed microelectrode had a sunflower shape. 1 - 5 μm of petal-shaped structures were formed along the circumference of the microelectrode and 500 nm of round-shaped grains were also formed at the center. Each structure was composed of numerous nanograins. The gold nanograin microelectrodes showed 56-fold decrease of interfacial impedance and 15-fold increase in charge injection limit compared to unmodified flat gold microelectrodes. By using dissociated rat hippocampal neuronal cultures, the recording and stimulation performance of gold nanograin microelec-trodes was tested. Noise level was 2.89 μVrms, which was low enough to detect low-amplitude neural signals (18.6 - 315 μV). Biphasic current pulses were applied to stimulate neurons and action potentials were evoked from neurons nearby electrodes. The developed nanograin neural electrode can be utilized to fabricate cell-based biosensors or clinical neural prosthetic devices.
전기 신호는 신경계에서 정보를 전달하는 중요한 매체이다. 신경 네트워크를 연구하려면 신경전자 인터페이스가 중요하다. 금속 미세전극은 신경계를 기록하고 자극하는데 널리 쓰인다. 미세전극의 성능는 조직-전극 인터페이스의 임피던스가 낮고 전자주입량 한계가 높을 수록 좋다. 미세전극의 성능을 향상시키기 위해 나노수준의 표면 공정이 연구되었다. 본 연구에서는 특이한 나노 금알갱이 구조를 표면에 가진 새로운 형태의 미세전극을 미세전극을 개발했다. 나노 금알갱이 미세전극은 해바라기보양으로 길이 1 - 5 μm의 꽃잎 구조가 전극 가장자리에 형성되었고 지름 500 nm의 구형 구조가 전극 가운데에 형성되었다. 각 구조는 수십 나노 크기의 나노 알갱이로 구성되었다. 나노 금알갱이 미세전극은 편평한 금 미세전극에 비해 임피던스는 56배 줄어들고 전하주입량은 15배 증가했다. 쥐의 해마세포를 배양하여 전극의 기록과 자극 성능을 테스트했으며 그 결과 배경잡음은 2.89 μVrms로 18.6 - 315 μV의 신경 신호가 측정되었으며 이상형 펄스를 가하여 반응 스파이크를 관찰할 수 있었다. 개발된 나노 금알갱이 미세전극은 바이오센서나 신경보철 분야 연구에 활용될 수 있을 것으로 기대된다.
