Extracellular matrix (ECM) in the stromal compartment modulates the growth and differentiation of mammary epithelial cells during embryonic and postnatal development. ECM of high stiffness has been shown to induce normal breast epithelial cells to have malignant phenotype with enhanced focal adhesion formation and reorganized actin stress fibers. Since actin stress fibers are in charge of intracelluar tension, the internal tension built up by ECM stiffness is suggested as a key regulator that induced phenotypic changes in cells.
To investigate the effects of externally applied mechanical tension on the phenotypic changes in normal breast epithelial cell line (MCF10A), we have developed a stimulator that applies cyclic uni-axial tensile stress on cells to induce internal tension build up inside cells. Upon 10%, 0.5Hz cyclic uni-axial tensile stimulation for 3 days, MCF10A showed significant increase in both mRNA and protein expression of matrix metalloproteinases-9(MMP-9), which is one of the key indicators for tumor invasion and metastasis. Using pharmacological inhibitor, Y-27632, Cytochalasin D and Latrunculin B, we show that actin polymerization is crucial for the uni-axial tensile stress induced regulation of MMP-9. We also observed significant increase in cellular motility, proliferation rate as well as irregular multilayer growth in cells that have been stimulated by uni-axial tensile stress. Our result may provide a key in understanding the role of cyclic tensile stress as an internal tension inducer, like in the case of stiff ECM, leading to malignant phenotypic changes in normal breast epithelial cells.
암은 지금까지 일반적으로 유전적 질환으로 이해되어 왔고 암세포의 모든 생물학적 특성이 암세포의 유전자(DNA)의 변화에서 기인한다고 알려져 있었다. 하지만 최근 유선상피에서 기원한 암의 발생이 세포외기질(Extracellular matrix, ECM)의 강성 증가와 연관성이 있음을 밝혀 주목을 받고 있다. 이와 같이 세포가 자라는 환경, 즉 세포 외기질의 강성은 세포 내부의 세포 골격(cytoskeleton)을 통해 내부로 전달되어 세포 내부 인장 (intracellular tension)을 조절하여 세포의 운동성과 분열에 영향을 준다. 본 연구는 세포외기질의 강성 증가를 통한 세포내부 인장 유도가 아닌, 세포 밖에서 직접적으로 인장 자극을 가하여 세포 내부의 인장을 효과적으로 증가시켜 정상 유선 상피세포인 MCF10A의 암적 성질 유도에 어떠한 영향을 미치는지 연구하였다.
본 실험은 PDMS membrnae위에 세포를 키우고, 단축 인장 자극기를 이용하여 세포에 인장자극을 주었다. 인장자극은 10% strain, 0.5Hz로 30분간 자극을 주고 30분간 휴지기를 주는 cycle을 반복하여 총 자극 시간이 3일이 되도록 실험하였다. 실험결과 실제 암의 발달 과정에서 기저막 분해에 핵심적인 역할을 하는 MMP-9 발현이 인장 자극에 의해 크게 증가하는 것을 mRNA와 단백질 레벨에서 확인하였고, MMP-9 발현은 세포내골격 단백질인RhoA-ROCK 신호전달을 통해 증가하는 것을 확인하였다. 또한 외부에서 가해준 인장 자극에 세포 내 p38 MAPK 신호전달을 활성화시키는 것을 관찰하였다. 인장 자극에 의해 세포가 무분별한 증식을 하여 다층을 이루는 것을 확인하였고, 자극이 가해지지 않은 세포에 비해 높은 운동성을 가지는 것을 정상 세포가 인장 자극에 의해 암적인 성질을 가지게 되었다는 것을 증명하였다. 암이 발생하고 진행 된 후 몸의 다른 장기로 퍼지게 되는 과정을 암의 전이라고 하는데, 이 과정에서 세포간 결합이 강하며 정적인 상피세포가 세포간 결합이 약하고 이동성이 높은 간엽세포로의 형질 변환이 일어난다. 외부에서 가하는 기계적 인장자극에 의해 정상유선상피세포의 상피-간엽 형질변환이 유도되는지 확인해보았으나, 인장 자극에 의해 형질변환이 일어나는 것으로 보이지 않았다. 이는 외부 인장자극에 의해 정상유선상피세포의 암적 성질은 유도되었으나 전이성을 가지게 되지는 않은 것으로 해석할 수 있다.
본 연구는 기존의 유전자 및 단백질간의 상호 신호전달 메커니즘 규명에 머물던 암 발생 연구의 확장으로, 세포 내의 기계적인 기작과 외부의 자극과의 직접적 연관성을 연구하였다는 점에서 큰 의미가 있다. 또한 암 세포의 발생 및 성장의 새로운 이해방법을 제시함으로써 현대사회의 중요한 질병인 암을 이해하는데 적극 활용될 수 있을 것이라 기대한다.
