Type I collagen is a major extracellular matrix component and its hierarchical structure plays an essential role in the regulation of cellular behavior. Here, I have analyzed the changes in the morphological, chemical, and mechanical properties of collagen fibrils induced by acidic and thermal treatments and the influence on the cellu-lar response of MC3T3-E1 cells. Morphological changes induced by the disintegration of the fibrillar structure of collagen were observed using atomic force microscopy. The changes in the surface chemistry due to the disas-sembly of native collagen fibrils were observed using time-of-flight secondary ion mass spectroscopy (ToF-SIMS). ToF-SIMS spectra were very sensitive to changes in the molecular configuration of the collagen fibrils induced by acidic and thermal treatments due to the extreme surface specificity. In addition, ToF-SIMS showed clear and reproducible changes in the surface amino acid composition corresponding to the acidic and thermal treatments of collagen fibrils. Based on the quantitative map of surface elastic modulus measured by contact-resonance force microscopy, acid and thermally treated collagen showed a lower elastic modulus than native collagen fibrils. Compared with native collagen fibrils, reduced cell spreading and decreased viability of MC3T3-E1 cells were observed on both the acid and thermally treated collagen.
Bone is a hierarchically organized biocomposite of organic (collagen) and inorganic (calcium phosphate, mostly hydroxyapatite) materials. The precise structural dependence and chemical correlation between mineral and collagen fibril are critical to unique characteristics of bone such as resilience and strength. Therefore, in ad-dition to the study of structural, chemical, and mechanical properties of collagen fibril, initial growth stages of hydroxyapatite was investigated using medium energy ion scattering spectroscopy (MEIS) for the understanding of bone formation mechanism. The quantitative compositional profiling with sub-nanometer resolution was con-ducted for hydroxyapaptite nanocrystals using MEIS.
본 학위 논문에서는 산 (acid) 그리고 열적 (thermal) 처리에 의한 콜라겐의 구조적, 화학 조성적, 기계적 특성의 변화를 표면 분석 장비인 원자힘현미경 (AFM), 비행시간이차이온질량분석법 (ToF-SIMS) 그리고 접촉 공진 힘 현미경 (CR-FM) 을 이용하여 연구하였다. AFM 을 이용하여 산, 열적 처리시의 콜라겐 섬유 각 가닥의 해체되는 과정에서의 구조적 차이점을 관찰하였고 ToF-SIMS 를 이용하여 콜라겐 표면의 아미노산의 조성 변화를 분명하고 재현성 있게 구분할 수 있음을 확인하였다. 또한 CR-FM 을 이용하여 산, 열처리시의 콜라겐의 탄성계수 변화를 이미징 하였고, 두 종류의 처리 후 콜라겐의 탄성계수는 모두 감소함을 보였다. 처리 후의 콜라겐 위에서 세포의 형상 및 생존율이 감소함을 보임으로서 산, 열처리후의 콜라겐의 구조적, 화학적, 기계적 특성변화가 세포의 접착 및 생존에 좋지 않은 영향을 미침을 확인하였다.
상호보완적인 AFM, ToF-SIMS, 그리고 CR-FM 을 이용한 콜라겐의 분해에 관한 구체적이고 자세한 연구는 조직공학 및 재생의학 분야에서의 콜라겐 사용에 있어 유용한 정보를 줄 수 있을 것으로 예상된다. 특히 본 연구의 결과는 콜라겐을 기반으로한 지지체의 기능성 평가 그리고 생물학적 지지체 제조시에 적용되는 세포 제거술 (decellularization) 프로세스 시에 일어나는 세포외 기질 (extracellular matrix)의 손상을 최소화 시키는 데 도움을 줄 수 있다.
