Optical imaging techniques are widely used in industrial and research fields: semi-conductor/FPD in-dustries, bio/medical and surface/material sciences, etc. However, optical imaging has an inherent barrier of resolution, limited by diffraction characteristic, in spite of numerous demands for high resolution. Various researches have been carried out to break the barrier (diffraction-limit barrier). Stimulated emission depletion (STED) microscopy, point spread function (PSF) engineering, and structured illumination microscopy (SIM) are introduced representatively. STED microscopy which depends on the energy transition phenomena has a limited application area related to fluorescent imaging. The techniques commonly require cumbersome procedures and elaborate arrangements. They have a poor axial resolution conventionally, because lateral resolution is enhanced at the expense of axial information.
In this thesis, point-scanning structured illumination microscopy (PSIM) is proposed to improve three-dimensional optical resolution concurrently. The resolution enhancement techniques of structured illumination are applied on point-scanning platform with computational acquisition algorithm. The light distribution signals for an object illuminated by a scanning spot are accumulated selectively and periodically during a scanning process to compose structured illumination images. The lateral and axial high frequency contents beyond the pass-band of OTF are obtained from the structured illumination images. The final image is reconstructed through superposition of the contents in spatial frequency domain. As a result the resultant OTF is extended along the three-dimensional direction, which induces the resultant PSF with a narrowed width. This result guarantees the imaging result of PSIM with improved optical resolution and depth dis-crimination.
PSIM has a huge advantage to generate an illumination pattern through a scanning spot moved easily and freely, not with additional external modulation but with simple computational algorithm. Structured illu-mination of PSIM is free from the fixed condition of the conventional structured illumination such as the illumination form and pattern period. The accumulation functions with a phase shift and different pattern period are determined to be suitable for a diffraction-limited spot illumination. Structured illumination images with phase shift for reconstruction, two-dimensionally structured illumination images for laterally isotropic enhancement, and structured illumination images of larger period for axially high frequency contents are obtained more easily and appropriately than the conventional SIM.
The working principles of PSIM are explained in comparison with that of the conventional SIM. A point-scanning based system configuration, an image acquisition process through periodic accumulation, and a reconstruction algorithm for structured illumination images based on spot-illumination are analyzed. The optical system is similar to the scanning system of the conventional confocal scanning microscope except for the detector. The point detector is replaced by an array detector to obtain the distribution information of an object illuminated by a focal spot. The structured illumination images are obtained by detecting the light dis-tribution signals periodically and accumulating according to the position of the focal spot. The contents for lateral and axial high-frequency are solved from two-dimensionally structured illumination images with a changeable spatial period; the final image is reconstructed from the contents.
The imaging process and result of PSIM are analyzed on the basis of wave-optical diffraction, to verify the feasibility of the proposed algorithm of PSIM theoretically. The light distribution at object plane and detector plane are derived, related to the illumination and the detection during a scanning process. The structured illumination image acquisition through the accumulation algorithm of the light distribution, and image reconstruction algorithm from the frequency contents of the structured illumination images are also analyzed mathematically on the basis of the aforementioned principles. The accumulation function of PSIM is determined appropriately according to the objective for resolution enhancement in three dimensions.
The numerical images for specific target objects are obtained through an image acquisition simulation following the imaging theory. The imaging performances of PSIM, as regards the resolution enhancement, are examined quantitatively from the estimated images, in comparison with those of other microscopy. The three-dimensional point resolution for a point object, the edge response for a lateral edge, the axial response for a flat object, and the resolving power for a patterned object are dealt with. OTF (optical transfer function) of the increased cut-off frequency, IPSF (intensity point spread function) of the narrowed width, and other improved results of PSIM are verified.
The optical system, including especially relay lens and imaging lens, is designed to satisfy the diffrac-tion-limit condition and their requirements. The relay lens satisfying the 4-f double-telecentric condition links the apertures of a mirror scanner and an objective lens to transfer scanning rays between them. The configuration and the variables of the relay lens are optimized considering design wavelength, beam diameter, magnification factor, and scanning angle. The imaging lens magnifies the light distribution on the small area of a spot-illuminated object to form its image at the detector plane. The configuration and the variables of imaging lens are established and optimized with consideration for the magnification factor required to fit the detector specification.
The experiments are conducted to evaluate the proposed imaging performances of PSIM. As a feasibility test for a scanning-spot based illumination, a two dimensional structured illumination pattern is confirmed at the object plane. The final reconstructed images for specific target objects are obtained through a bright-field imaging experiment. The imaging performances including one-point resolution, two-point resolution, edge response, pattern expressivity, axial response, and three-dimensional image are examined and compared with the data expected by the numerical simulation.
In conclusion, the three-dimensional resolution enhancement of PSIM is verified theoretically, numer-ically, and experimentally. PSIM can investigate a sample more in detail and three-dimensionally from the reconstructed images with sub-diffraction resolution. The advantages of PSIM increase the utility of SIM and improve spatial resolution in three dimensions through two-dimensional structured illumination with change-able periods. In the future, PSIM will be upgraded for an additional improvement by applying to PSF engi-neering or to 3D structured illumination; its application area will be extended to nonlinear imaging with high resolution.
광학 이미징 기술은 산업 현장 (반도체, 평면디스플레이 기기 생산) 과 연구 분야 (생체 의료 연구, 표면 및 재료 연구) 에서 영상을 통한 검사/측정/분석 도구로서 널리 사용되어 왔다. 해당 분야의 관찰 대상 크기는 작아지고 있고 보다 자세하게 분석하기 위하여 높은 분해능을 가진 영상을 필요로 함에도 불구하고, 광학 이미징 기술에는 회절 현상으로 인한 분해능을 제한하는 장벽이 존재한다. 회절 한계 분해능 장벽을 깨고, 고분해능 광학 이미징을 구현하기 위하여 다양한 연구가 시행되어 왔다. 형광 물질에 의존하지 않고, 복잡한 처리 절차와 정교한 정렬 수준으로부터 자유로우며, 횡방향 분해능 향상뿐 아니라 축방향 분별력도 갖게 하여 3차원 이미징도 가능한 광학 이미징 기술로서, 점주사 구조 조명 현미경을 본 논문에서 제안한다.
기존의 구조 조명 현미경의 기본 분해능 향상 기법을 기반으로 하여, 점주사 시스템과 주기적 영상 누적/획득 기법을 적용하였다. 초점 분포를 통해 물체를 조명하고, 점주사 과정동안 움직이는 초점의 주기적인 위치에 대하여, 광분포 신호들을 선택적으로 누적시킴으로써 구조 조명 영상을 획득한다. 기존의 구조 조명 현미경과 같이 정해지지 않은 구조에서 추가적인 변조 장치 없이 간단한 전산 알고리즘을 통해서, 조명 패턴을 쉽고 자유롭게 생성한다. 영상 복원을 위해 위상차를 갖는 구조 조명, 등방적 분해능 향상을 위한 2차원 구조 조명, 광축 정보를 획득하기 위해 주기가 조절되는 구조 조명을 생성하고 영상을 획득한다. 획득된 구조 조명 영상들로부터 일반 광학 현미경의 광학 전달 함수 너머의 횡방향 고주파수 성분, 축방향 성분들을 구하고 이를 공간 주파수 분포에서 더하여 복원함으로써, 3차원적 분해능이 향상된 최종 영상을 획득한다. 결과적으로 제안된 점주사 구조 조명 현미경의 광학 전달 함수는 3차원 방향으로 넓어지고, 점상분포함수는 폭이 좁아진다.
점주사 기반의 광학 시스템 구성과 주기적 누적법 기반의 영상 획득 절차, 초점 조명 기반 구조 조명 영상의 복원 알고리즘을 제안하고 분석하였다. 광학 시스템은 점광검출기 구조만 제외하고 공초점 현미경의 점주사 시스템과 유사하다. 기존에 존재하던 광선의 각도를 조절하여 초점 조명을 주사하는 구조에서, 영역 광검출기를 사용하여 초점 조명 주변의 물체 영역 정보를 획득한다. 선택적으로 광분포 신호들을 획득하고 점주사 위치에 따라 누적함으로써 구조 조명 영상을 획득하고 최종 영상을 복원하는데 활용한다.
점주사 구조 조명 현미경의 영상 획득 과정과 결과에 대해 제안된 원리를 회절 이론을 기반으로 분석하고 검증하였다. 점주사 과정 중에서 시간에 따라 변화하는, 물체 평면에서의 조명 광분포 정보를, 광검출기 평면에서의 광분포 정보를 분석한다. 검출 광분포 정보를 설계된 누적 함수를 따라 연산 처리함으로써 초점 조명 기반의 구조 조명 영상을 표현한다. 위치 이동 되어 구조 조명 영상에 포함된 횡방향 고주파수 성분을 확인하고, 초점 조명에 맞도록 영상 복원 알고리즘을 설계하며 이에 대한 최종 복원 영상을 표현한다. 누적 함수는 3차원 분해능 향상 및 2차원 분해능 최대 향상을 위한 조건에 따라서 적절하게 설계하였다.
이미징 이론을 따르는 영상 획득/복원 알고리즘을 적용하여, 특정 물체에 대한 영상을 수치적인 시뮬레이션을 통해 획득하였다. 분해능 향상과 관련된 이미징 성능을 예측하고, 타현미경의 이미징 성능과 비교한다. 향상된 임계 주파수를 가지는 광학 전달 함수, 폭이 좁아진 3차원 한 점 분해능, 날카로워진 횡방향 모서리에 대한 응답 특성, 횡방향 패턴 물체에 대한 향상된 분별력, 축방향 응답 특성을 따른 깊이 분별력 등을 시뮬레이션을 통해 검증하고 수치적으로 확인한다.
광학 시스템을 시스템 요구사항과 회절 한계 성능에 맞도록 설계하였다. 4-f 더블 텔레센트릭 구조의 중계 렌즈는 주사 거울로부터 각도가 조절된 점주사 광선을 대물렌즈로 전달한다. 긴 초점 거리의 이미징 렌즈는 초점 분포에 의해 조명된 물체 작은 영역을 광검출기의 큰 픽셀들의 중심에 각각 할당되도록 확대된 상을 형성한다. 설계 파장, 광선 직경, 점주사 각도를 고려하여 중계 렌즈가 설계되었고, 획득 영상에 요구되는 픽셀 수, 광검출기 픽셀 크기에 맞도록 광학계 확대율 및 초점 거리가 결정되어 이미징 렌즈가 설계되었다.
설계된 결과를 바탕으로 시스템을 제작하여 점주사 구조 조명 현미경을 구성하였다. 물체 평면에서 점주사 초점 기반의 2차원 구조 조명 패턴을 직접 살펴봄으로써 구조 조명의 가능성을 따져 보았다. 일반 광을 이용하여 특정 물체에 대한 영상 획득 실험을 수행하여 공간 분해능 향상을 검증하였다. 앞서 수치적인 시뮬레이션을 통해 살펴보았던 항목들을 실험적으로 검증하였고 수치적인 예상 결과값과 비교하였다. 한 점 분해능, 두 점 분해능, 모서리 응답 특성, 패턴 물체 분별력, 광축 응답 특성, 3차원 높이 영상 획득과 관련된 이미징 성능의 향상 정도를 실험적으로 확인하였다.
요약하자면, 점주사 구조 조명 현미경의 3차원 분해능 향상 원리를 제안하였고, 시스템을 설계하여 구성하였으며, 이미징 성능에 대해서 이론적으로, 수치적으로, 실험적으로 검증하였다. 기존의 구조 조명 현미경에서 제한된 응용성을 점주사 방식을 통해 증대시키고, 주기 조절이 가능한 2차원 구조 조명을 통해 3차원 분해능이 향상된 영상을 획득하였다. 회절 한계 이하의 횡방향 분해능, 축방향 분별력을 통한 3차원 광학 영상을 통해 미세 패턴, 조직 등을 가지는 물체들을 보다 크게 확대하여 자세하게 분석할 수 있을 것이고 이는 분석 결과의 신뢰성을 증대하는데 기여할 것이다. 앞으로 점상 분포 함수 조절 기법이나 3차원 구조 조명 생성에 점주사 방식이 효과적으로 적용되어 점주사 구조 조명 현미경의 횡방향, 축방향 분해능 성능은 더욱 향상될 것이고, 점주사 및 이미징 처리 속도가 개선되어 영상 획득 속도가 향상될 것이다. 또한 점주사 방식만이 사용 가능한 비선형 광학 기반의 광학 이미징 분야에 적용되어, 이에 대한 고분해능 영상 정보를 얻는데 응용될 것이다.
