This thesis presents a novel immunoassay platform based on the optoelectrofluidic mobility of immunocomplexes, in which analyte-bound microbeads are conjugated with secondary antibodies with gold nanoparticles. Recently, optoelectrofluidics, the technology which is based on the electrokinetic motions of particles or fluids under a light-induced electric field, has become a powerful technology for manipulating microparticles and biological materials such as cells and molecules, and for detecting disease biomarkers. In this paper, we propose a new optoelectrofluidic sandwich immunoassay method using difference of immunocomplexes’ mobility which is determined by the amount of captured gold nanoparticles for detecting the concentration of target antigens. Here, simulations for electric field distribution, Clausius??Mossotti factors and net force affecting on the immunocomplexes are carried out in order to verify feasibility the principle of this study. Besides, we demonstrate the sandwich immunoassay for detecting murine IgG with gold nanoparticles which have different sizes such as 5 and 10 nm. The velocity of immunocomplexes is proportional to the concentration of IgG and also depends on the size of gold nanoparticle used. This result implies that we can easily control the detection sensitivity of immunoassay by controlling the size of gold nanoparticle. The sandwich immunoassays using different IgGs (murine IgG and human IgG) with the same-sized gold nanoparticle (5 nm) are also carried out. Despite the same-sized gold nanoparticles, the mobility is different. From the results, it seems that the different mobility might result from intrinsic affinity properties of target antigens.
본 연구에서는 간단하고 빠르게 면역분석법을 수행하기 위한 새로운 광전자유체 (Optoelectrofluidic) 샌드위치 면역분석법을 제안하였다. 최근에는 적은 양의 시료로도 고감도의 면역분석을 빠르고 정확하게 수행할 수 있는 미세유체 기반 기술의 중요성이 대두되고 있으며, 특히 미세입자를 이용한 면역분석법의 경우에는 입자 표면을 처리하는 다양한 기술이 이미 상용화되어 있고 자유자재로 입자를 조작할 수 있기 때문에, 면역분석을 위한 랩온어칩 (Lab-on-a-Chip) 개발의 차세대 후보로 주목 받고 있다. 한편, 지금까지 개발된 미세입자를 이용한 미세유체 면역분석 기술은 복잡한 측정 시스템이 필요하거나 세밀한 조작이 어렵다는 한계를 가지고 있기 때문에, 간단한 시스템만으로도 정교한 조작과 측정을 수행할 수 있는 기술이 필요하였다.
본 연구에서는 액정디스플레이 (liquid crystal display, LCD) 기반의 광전자유체 시스템을 이용하여 미세입자를 자유자재로 조작 할 수 있을 뿐만 아니라, 미세입자의 운동성 변화를 측정하여 항원의 농도를 검출하기 때문에 복잡한 측정 시스템이 필요치 않고 아주 간단히 면역분석을 수행할 수 있었다. 특히, 본 연구에서는 시뮬레이션을 통하여 제안한 새로운 면역분석법의 원리를 수학적으로 검증하고, 두 종류의 항원 (human IgG, murine IgG) 과 크기가 다른 금 나노입자 (5 and 10 nm) 를 이용하여 항원의 종류와 금 나노입자의 크기가 본 기술에 미치는 영향을 연구하였다. 이를 통하여, 면역분석을 간단한 시스템 하에서 고속으로 수행함과 동시에 in situ 와 on-demand 한 조작을 할 수 있는 본 기술의 장점을 검증할 수 있었으며, 이는 고효율의 분석을 위한 타 기술과의 집적화가 가능할 뿐 아니라, 가상전극 (virtual electrode) 을 이용하여 입자를 자유자재로 조작할 수 있기 때문에 다중면역분석이 가능한 통합 플랫폼 개발에 이용될 수 있을 것이라 사료된다.
