Synchronization is a collective phenomenon occurring in systems of interacting units and is ubiquitous in neural system. But failure of this feature, abnormal synchrony, can be occurred in brain disease such as schizophrenia. Recent studies have enlightened the abnormalities of beta and gamma frequency band in the brain of schizophrenia patients which are closely related with specific symptoms of schizophrenia. However, most of these studies neglect the mechanism of such abnormalities. Here, we analyzed feedback network topology composed of MEG magnetometer sensors and investigated topological differences between normal and schizophrenia patients group during resting condition. The number of feedback circuits were differentially enriched in two groups representing weaker positive feedback interaction within brain cortical region, local feedback, at patient. At this moment, we proposed the minimal structure, coupled positive-positive feedback (CPPFB), existing in their network. In terms of simulation results using Wilson-Cowan model, CPPFB structure showed decreasing beta band phase locking value and changing dominant frequency power to gamma range. Schizophrenia patients showed lack of this structure in their network and therefore related phenomena were significantly reduced at patients group. Finally, severe negative symptoms were caused for those who has less CPPFB structure in their network in schizophrenia patients.
뇌 신호의 동기화 현상은 신경세포들간의 상호작용에 의한 결과이고 인간의 인지기능을 포함한 대부분의 뇌 기능을 대변하는 현상이다. 특히 정신분열병은 인지기능의 저하와 관련해 신경세포간 동기화에 실패하는 대표적인 뇌 질환으로 간주되는 만성 질병이다. 최근 수 년 동안 뇌 신호 측정기술의 발달에 힘입어 정신분열병 환자의 신경세포간 동기화가 정상적으로 이루어지지 않는다는 사실이 많은 연구를 통해 밝혀져 왔지만, 대부분의 연구들이 신경세포들 간의 상호작용에 대한 생물학적인 현상만을 보고할 뿐 그 기저가 되는 근본원인은 제시하지 못하고 있다. 본 연구는 이러한 현상의 핵심 원리를 규명하고자 뇌자도 신호를 분석하여 정상군과 정신분열병 환자군의 휴지기 신경네트워크를 추론하였고, 신호 발생의 원인이 되는 신경네트워크 핵심 구조를 제시하였다. 바로 CPPFB구조는 서로 다른 뇌 피질 영역간의 상호작용과 동일한 피질 영역 내에서의 상호작용이 서로 긴밀하게 결합된 구조인데, 이러한 기본 구조가 정신분열병 환자의 신경네트워크에서 감소하였으며, 이는 정신분열병의 음적 증상과도 유의미한 관계가 있다는 것이 본 연구에서 밝혀졌다. 그 동안 정신분열병 증상에 관한 어떠한 근거도 제시된 적이 없었던 점으로 미루어 볼 때, 본 연구의 결과는 앞으로 정신분열병의 원인이 뇌 피질 신경세포들간의 상호작용 결여에 의한 것이라는 가능성을 제시할 수 있다.
