A new platform for lab-on-a-chip system is suggested that utilizes a biosensor array embedded in a digital microfluidic device. With field-effect transistor (FET)-based biosensors embedded in the middle of droplet-driving electrodes, the proposed digital microfluidic device can electrically detect avian influenza an-tibody (anti-AI) in real time by tracing the drain current of the FET-based biosensor without a labeling pro-cess. Digitized transport of a target droplet enclosing anti-AI from an inlet to the embedded sensor is enabled by the actuation of electrowetting-on-dielectrics (EWOD). A reduction of the drain current is observed when the target droplet is merged with a pre-existing droplet on the embedded sensor. This reduction of the drain current is attributed to the specific binding of the antigen and the antibody of the AI. The proposed hybrid device consisting of the FET-based sensor and an EWOD device, built on a coplanar substrate by monolithic integration, is fully compatible with current fabrication technology for control and read-out circuitry. Such completely electrical manner of inducing the transport of bio-molecules, the detection of bio-molecules, the recording of signals, signal processing, and the data transmission process does not require a pump, a fluidic channel, or a bulky transducer. Thus, the proposed platform can contribute to the construction of an all-in-one chip.
본 연구에서는 선충전 (Pre-charging) 방식의 물방울 구동법과 새로운 랩온어칩의 플랫폼으로서 디지털 마이크로플루이딕 소자 내에 바이오센서를 탑재한 시스템을 제안한다. 선충전 방식의 물방울 구동법은 Electrowetting-on-dielectrics (EWOD) 기반의 단일 평판 구조 디지털 마이크로플루이딕 소자에 적합한 구동 방식으로써, 물방울을 움직이기 전에 물방울을 선충전 시킴으로써 기존의 Electrowetting에 의한 힘에 정전기적 인력을 더해 보다 큰 구동력으로 물방울의 움직임을 가능하게 한다. 따라서 보다 낮은 전압을 이용하고도 효과적으로 물방울을 움직일 수 있다는 장점이 있다. 본 연구에서 제안하는 바이오센서를 탑재한 디지털 마이크로플루이딕 소자는 디지털 마이크로플루이딕 소자의 물방울 구동을 위한 전극의 한 가운데에 전계 효과 트랜지스터를 기반으로 한 바이오센서를 두고, 이 바이오센서의 드레인 전류를 측정함으로써 조류독감 항체를 전기적인 방법을 통해 실시간으로 검출할 수 있다. 이러한 방법은 복잡한 레이블링 과정 (Labelling process)을 거치지 않고도 특정 바이오 물질을 검출해 낼 수 있다는 장점이 있다. 선충전 방식의 물방울 구동법을 적용하여 조류독감 항체 등의 바이오 물질이 들어있는 물방울들을 이동하였으며, 조류독감 항체가 들어있는 물방울이 바이오센서 위의 기존의 물방울과 합쳐지자 드레인 전류가 감소하는 것을 확인하였다. 이는 물방울 안에 들어있던 조류독감 항체가 바이오센서 표면에 고정되어있던 조류독감 항원과 특정 결합 (Specific binding)을 하게 되면서, 조류독감 항체의 음(-)전하가 n-channel 트랜지스터로 이루어진 바이오센서의 전류 흐름을 방해하였기 때문이다. 본 연구에서는 전계 효과 트랜지스터를 기반으로 한 바이오센서와 EWOD의 원리로 구동되는 디지털 마이크로플루이딕 소자를 하나의 기판 위에 집적함으로써, 차후 제어 및 Read-out 회로의 추가가 용이하도록 하였다. 이러한 개념의 소자는 하나의 칩 위에서 바이오 물질이 포함된 물방울을 움직여 실험을 수행하고, 바이오 센서를 통해 실험으로부터 얻어진 바이오 물질을 검출하며, 이 과정에서 얻은 신호 및 정보를 처리하여 전송까지 하는 모든 과정을 전기적으로 가능하게 하는 차세대 All-in-one-chip 소자의 개발에 응용될 수 있을 것이다.
