Production of enantiopure compounds is essential in the pharmaceutical industry. Among the various methods to synthesize or to separate desired optical isomer, enzymatic chiral resolution is promising technique due to its mild reaction condition and substrate specificity of the enzyme. Oxidoreductase is an enzyme that catalyzes oxidation or reduction of the specific chemicals. To act as a bio-catalyst, it needs specific cofactors, which are usually nicotinamide derivatives. Nicotinamide derivatives are consumed in those enzymatic reactions as a stoichiometric manner. Hence, it is challenging to improve enzymatic reactions into large scale due to the expensiveness of these nicotinamide derivatives.
To overcome this problem, we introduced light-driven cofactor regeneration system to the enzymatic chiral resolution. Cofactor regeneration system can recycle the enzymatic cofactors from the visible-light by the interactions of electron donor, photosensitizer and electron mediator.
In this study, we selected ketoreductase 130 enzyme to produce key precursor, (R)-2-chloro-1-phenylethanol, for the synthesis of the fluoxetine. This enzyme employs NADPH as its cofactor. As we employed light-driven cofactor regeneration system, we introduced NADP+ instead of NADPH. As a result, we could get the desired chiral product. This implies that our enzymatic chiral resolution via light-driven cofactor regeneration system worked well because we introduced NADP+ in this reaction.
Generally, nearly all the substrate was consumed in only 30 minutes after ‘light-on’ stage. Nevertheless, yield was low compared to the conversion value. Meanwhile, enantiomeric excess was perfect during the reaction. We successfully got the only desired chirality. To examine the effect of essential reagents in this reaction, we varied the concentration of Rh* complex (an electron mediator) and enzyme. From this experiment, we confirmed that main driving force of this reaction is enzymatic reaction, instead of cofactor regeneration system. Moreover, we discovered byproduct during the reaction by the HPLC analysis. The reason may due to the interaction between Rh* complex and substrate.
Finally, we varied the xanthene dyes to compare the effect with eosin Y which was main photosensitizer used in the reaction. As a result, we could assume that some of the transferred electron was used to reduce the NADP+. The portions of transferred electron were varied.
From all these results, we hopefully assume that our system can be utilized to the pharmaceutical industry, fine chemicals and agrochemicals due to its advantage over the conventional methods because of the reduced costs of the process.
현대의 제약산업에서 광학활성 의약물의 비중은 점점 증가하고 있고, 이에 따라 순수한 광학활성 의약물을 생산하는 과정과 공정은 최근에 많은 주목을 받고 있다. 이 중에서 효소를 이용하여 광학 이성질체를 생산, 분리하는 방법이 최근에 주목을 받고 있는데, 이는 효소반응의 광학적 기질특이성과 효소반응 자체가 가지는 과정의 용이함에 기인한 것이다. 이러한 효소 중에서 산화환원 효소는 다양한 물질들의 산화와 환원반응을 촉매하는 효소이다. 산화환원 효소는 생화학적 촉매반응을 수행하기 위하여 니코틴아미드 조효소를 필요로 하는데, 이 조효소는 일반적으로 가격이 비싸므로, 우리가 필요로한 물질을 대량생산을 하는데에 큰 단점으로 작용한다. 빛을 이용한 인공광합성 시스템은 이러한 니코틴아미드 조효소를 재생할 수 있는 시스템으로, 값비싼 니코틴아미드 조효소의 사용을 최소화 하여 원하는 생성물을 과량으로 얻는 효소반응을 충분히 수행할 수 있는 장점을 가지고 있다. 본 연구에서는 이러한 효소를 이용한 광학이성질체의 분리와 빛을 이용한 인공광합성 시스템 간의 장점을 합하여 산화환원효소 중의 하나인 ketoreductase 효소를 이용하여 항우울제의 일종인 fluoxetine의 합성과정의 중간물질인 (R)-2-chloro-1-phenylethanol을 생성하였다. 본 반응에서는 우리가 원하는 순수한 광학 이성질체뿐만이 아니라 다른 부산물도 형성이 되었는데, 이는 빛을 이용한 인공광합성 과정에서 반드시 필요한 유기금속촉매인 로듐복합체와 반응기질물질간의 상호작용이 일어나서 로듐복합체의 구조적 변화가 야기된 것이 주요한 원인으로 생각된다. 이러한 결과를 종합해 보았을 때, 빛을 이용한 인공광합성 시스템을 통한 광효소적 이성질체 분리반응의 주요 추진동력은 효소반응이며, 로듐복합체 자체가 광화학적 환원반응을 일으킬 수 있으므로 반응수율을 높이기 위해서는 효소와 로듐복합체 사이에서 적절한 반응비율을 이루어야 한다는 사실을 알게 되었다. 또한 크산텐 염료가 본 인공광합성 시스템에서 광민감제의 역할을 수행하는데, 위 실험에서 주로 사용된 크산텐 염료 중의 하나인 에오신 와이를 포함한 다양한 크산텐 염료를 사용하여, 반응의 속도와 이에따른 ketoreductase의 니코틴아미드 조효소인 NADPH 생성속도를 비교하였다.
우리의 이와 같은 연구는 앞으로 광학이성질체의 효소반응을 통한 생성과정에서 상당한 비용절감을 가져올 수 있는 기술로써, 앞으로의 상용화가 기대되는 연구라 할 수 있다.
