Proteins can be engineered toward better properties by directed evolution technologies with high throughput screening and, protein display system in which genotype and phenotypes are physically linked, is the most powerful tool for this purpose. So far, several display systems including phage display, cell surface display, etc have been developed and widely used in bacterial and yeast host cells. However, even though many proteins have been engineered successfully with those plasmid systems, each system has several drawbacks to be improved - particularly, no display systems are suitable for cytoplasmic proteins yet. In this study, we developed new protein display system, plasmid display system in which target protein is linked to plasmid DNA in cytoplasm of E. coli. As an anchoring motif to plasmid, DNA binding domain (DBD) of human Oct-1 protein was employed which can bind to specific DNA sequence (ATGCAAAT) with high affinity (KD = 9x10-11 M). With the GST fused model, the conditions for gene expression and isolation of protein-linked plasmid complex were optimized. Through three to four rounds of en-richment process, the GST tagged fusion proteins were successfully isolated from a protein mixture with a starting ratio of 1:102 and 1:104. The results demonstrate that Oct-1 based plasmid display system is suitable for the isolation of engineered proteins from huge libraries.
생명공학에서의 디스플레이 시스템은 특정 물질 (DNA, RNA, 다른 단백질 등등)에 기존의 라이간드 (항체, 효소, 폴리펩타이드 등등)보다 더 높은 결합력으로 결합할 수 있는 혹은 더 열역학적으로 안정된 재조합 단백질을 여러 번의 enrichment를 통하여 찾아내는 과정을 말한다. 이전의 수많은 논문에서, phage display와 cell surface display는 비교적 높은 library size (107-1010) 를 통하여 향상된 재조합 단백질을 찾는데 널리 사용되어 왔다. 그러나 스크리닝을 위해서는 periplasm 영역 혹은 세포 밖으로 재조합 단백질의 분비 과정이 필요하므로 모든 재조합단백질을 완벽히 스크리닝 하는데 한계가 있었다. 또한 mRNA 와 ribosome 디스플레이는 완벽한 in vitro 실험을 통하여 위의 한계점을 극복하고, 매우 높은 library size (1013) 를 구현하였다. 그러나 이 역시, 불안정한 mRNA를 사용하고, in vitro 상에서 translation을 유도하기 위하여, 세포 안에서의 조건과 유사하게 시스템을 제어해야 하기 때문에 기술적으로 민감할 수 있었다. 이러한 문제점을 해결하고, 주로 cytoplasmic 단백질을 스크리닝 하기 위하여 본 연구에서는 새로운 플라스미드 디스플레이 시스템을 개발하였다.
우선, 본 연구에서는 규모가 큰 단백질 라이브러리로부터 목적 단백질을 분리하기 위하여 oc-tamer sequence 와 Oct-1 POU domain 유전자를 포함하는 플라스미드 디스플레이를 제작하였다. 또한 시스템의 선택도를 향상시키기 위하여, 대장균에서의 유전자 발현조건과 in vitro 상에서의 단백질-플라스미드 복합체를 분리하는 조건을 최적화 하였다. 뿐만 아니라, Oct-1 POU domain과 DNA사이의 결합 구조를 분석하여, in vitro 분리과정에서 단백질-플라스미드의 복합체가 안정적으로 유지 될 수 있는 방안을 모색하였다. 그 결과 Oct-1 POU domain의 C-말단 보다 N-말단에 affinity tag을 융합하였을 때, in vitro 분리과정에서 단백질-플라스미드의 복합체가 보다 안정적으로 유지될 수 있음을 확인하였다. 또한 최적화한 조건으로 위의 plasmid system과 affinity tag이 없는 대조군 plasmid system과의 세포 혼합 비율을 각각1:102, 1:104으로 하여 enrichment를 진행하였다. 비록 적은 양의 세포규모에서 실행하였지만, 두 가지 경우 모두, 3~4회의 enrichment 과정 이내에서 affinity tag을 가진 plasmid system이 증폭되는 것을 확인할 수 있었다.
플라스미드 디스플레이 시스템에 관한 연구는 과거에도 있었지만, 실제로 대장균 내에서 재조합 단백질을 효율적으로 스크리닝한 사례가 적기 때문에 아직 이론적인 장점을 입증하는 단계에 머물고 있다. 그러므로 앞에서 언급한 것과 같이, 본 연구에서는 기존의 디스플레이 시스템의 단점을 보완하면서 cytoplasmic 단백질을 보다 효율적으로 스크리닝하기 위한 실용적인 시스템으로 만드는 것을 목표로 하였다. Enrichment 결과에서 보는 것과 같이, 단백질 혼합 용액 내에서 목적 단백질을 분리할 수 있음을 입증하였고, 이는 간단한 공정과정을 통하여 큰 규모의 라이브러리로부터 결합력이 뛰어난 재조합 단백질을 선택적으로 분리할 수 있는 가능성을 제시한 것이다.
