Tetrahydrobiopterin (BH4) deficiency is a genetic disorder associated with a variety of metabolic syndromes such as phenylketonuria (PKU). In this article, the biochemical mechanism by which BH4 deficiency inactivates mTORC1 leading to the activation of autophagic pathway was studied utilizing BH4-deficient Spr??/?? mice generated by the knockout of the gene encoding sepiapterin reductase (SR) catalyzing BH4 synthesis. In tissues from Spr??/?? mice, mTORC1 signaling was inactivated as assessed by the phosphorylation of S6K and autophagic pathway was elevated as assessed by the conversion of cytoplasmic LC3-I into lipidated LC3-II. This study has demonstrated that tyrosine deficiency causes mTORC1 inactivation and subsequent activation of autophagic pathway in Spr??/?? mice. Therapeutic tyrosine diet completely rescued dwarfism and mTORC1 inhibition but inactivated autophagic pathway in Spr??/?? mice. Tyrosine deficiency-dependent inactivation of mTORC1 in Spr??/?? mice was also supported by mTORC1 inactivation in Pahenu2 mouse model lacking phenylalanine hydroxylase (PAH). The phosphorylation of S6K was severely reduced and the conversion of LC3 was elevated in tissues from Pahenu2 mice. NIH3T3 cells grown under the condition of tyrosine restriction exhibited autophagy induction but high levels of phenylalanine had no effect on the activation of autophagic pathway. In addition, ectopic expression of RhebQ64L, a constitutively active mutant of Rheb, activated mTORC1 leading to suppress autophagic pathway even in the tyrosine deficient condition. This study also documents mTORC1 inactivation and autophagy induction in PKU patients with BH4 deficiency. Moreover, defective locomotive activities of Spr??/?? mice were almost completely rescued by tyrosine supplementation, suggesting that tyrosine deficiency is an important pathophysiological cause of symptoms associated with BH4 deficiency.
테트라히드로비옵테린 (BH4)의 결핍은 선천적 유전질환의 일종으로 페닐케톤뇨증 (PKU)를 비롯한 다양한 대사증후군을 수반한다. 본 연구에서는 BH4의 생합성에 관여하는 중요한 효소인 sepipterin reducatse (SR) 유전자를 녹아웃 시킨 Spr-/-??생쥐를 이용하여 BH4의 부족상황이 mTORC1 신호계를 억제하여 자가식작용(autophagy)를 활성화시킨다는 생화학적 메커니즘을 연구하였다. Spr-/- 생쥐의 조직에서 mTORC1 신호계가 억제되고 있음을 S6K의 인산화로 확인하였고, 지질화된 LC3-II의 생성여부를 통해서 자가식작용이 활성화 되고 있음을 확인하였다. 또한 이러한 현상이 티로신의 결핍에 의해 일어난다는 점도 제시하였다. 티로신을 Spr-/-??생쥐에 공급해 주면 Spr-/-??생쥐의 왜소증(dwarfism)이 회복되었고 mTORC1의 재활성화, 자가식작용의 억제가 유도되었다. 티로신에 결핍에 의한 mTORC1 신호계의 억제는 다른 PKU의 생쥐모델로서 페닐알라닌 수산화 효소(PAH)의 활성이 저하되어있는 Pahenu2 생쥐에서도 확인할 수 있었다. Pahenu2 생쥐에서도 S6K의 인산화가 현저히 감소되어있었고, LC3-II의 전환도 상당히 증가되어 있음을 확인하였다. NIH3T3 세포주를 이용한 실험에서는 고농도의 페닐알라닌은 자가식작용을 유도하지 않음을 확인하였다. 또한 Rheb의 활성형 돌연변이체인 RhebQ64L을 NIH3T3 세포주에 발현시키면 티로신이 결핍된 배지에서도 mTORC1의 활성이 유지됨은 물론 자가식작용도 유도되지 않음을 확인하였다. 본 연구는 또한 BH4 결핍증 환자의 림프구에서 mTORC1의 활성이 억제됨과 자가식작용이 유도되고 있음을 제시하였다. 더욱이 Spr-/-??생쥐의 행동장애가 티로신의 공급으로 대부분 회복됨을 보임으로서 BH4 결핍증과 관련된 병리학적 원인에 티로신 결핍이 중요함을 제시하였다.
