A series of supramolecular graft copolymers (SGPs) composed of $Poly(_L-lysine)$ (PLL) as a main chain and $poly(_{D,L}-lactic-co-glycolic acid)$ (PLGA) as a side chain was synthesized with different degrees of substitution (DSs) through ionic interaction in a DMSO solvent. The addition of water induced the self-assembly of PLL-PLGA SPGs into spherical micelles at a critical water content (CWC).
The size of the spherical micelles at each CWC decreased with an increase of DS and core-shell structures, which was confirmed by XPS and zeta-potential measurements. The morphological transitions were driven under thermodynamic control when the solvents mixed.
Consequently, further increases of water content caused the transition of the spherical mi-celles to vesicles, while the initial spherical micelles were reconstructed from vesicles when the DMSO was added. The content of water, the boundaries for the morphology transition, depended on the DS.
The pH and salt levels affected the stability of the aggregates weakening the ionic interactions between the PLL and PLGA.
본 연구에서는 PLL-PLGA 초분자 그라프트 공중합체를 합성 한 후, 자기조립을 유도하여 자기조립체의 구조변이와 물리, 화학적 특성을 연구하였다.
DMSO용매 상에서 양 이온성 친수성 고분자인 $Poly(_L-lysine)$ (PLL)에 이온결합을 이용하여 소수성인 $poly(_{D,L}-lactic-co-glycolic acid)$ (PLGA) 사슬을 접목시켜 4가지 서로 다른 사슬 접목도를 갖는 양친성 초분자 그라프트 공중합체를 합성하였다.
용매의 조성과 PLGA 사슬 접목도에 따른 PLL-PLGA 초분자 그라프트 공중합체의 구조변이를 관찰하기 위해 각각 DMSO 용매에 용해된 1.6, 3.8, 6.9, 14.2%의 사슬 접목도를 지닌PLL-PLGA 초분자 그라프트 공중합체에 천천히 증류수를 첨가하며 교반 시킨 결과, PLGA 사슬 접목도 증가에 따라 각각 32, 20, 16, 4%의 증류수/DMSO 용매조성에서 자기조립체를 형성하는 것을 UV/vis spectroscopy로 확인하였다. 각각의 CWC에서 형성된 자기조립체의 구조와 물리, 화학적 특성을 TEM, DLS, X-ray Photoelectron Spectroscopy (XPS), Zeta-potential Measurement를 이용하여 측정한 결과 spherical micelle 구조를 가지며 PLGA 사슬 접목도 증가에 따라 크기가 작아지는 것을 관찰하였다.
증류수를 더욱 첨가하여 증류수/DMSO용매에 용해된 14.2%의 PLGA사슬 접목도를 지닌 자기조립체의 용매조성을 변화시킨 결과 spherical micelle구조에서 vesicle구조로 변화하는 것을 관찰 할 수 있었다. 다시 DMSO를 첨가하여 초기의 증류수/DMSO조성으로 변화시킬 경우 vesicle 구조에서 spherical micelle 구조로 전이하는 것을 관찰 할 수 있었다.
마지막으로 자기조립체의 pH와 염에 대한 자극감응성을 관찰하였다. 이것은 PLL과 PLGA 사이의 이온결합의 붕괴에서 비롯된 결과라고 해석할 수 있었다.
