Various shapes of micropatterns printed on the surface have been used to could control cellular behaviors, such as migration and division, in vitro. A neuron has unique cellular behaviors, such as neuritogenesis and axonal protrusion, which play an important role in the connectivity of neuronal circuitry. Here we designed micropatterns for controlling these behaviors of neurons. Ten types of micropatterns were designed with about 130 $μm^2$ of area for controlling lamellipodia surrounding the spherical cell body. All micropatterns composed of the mixture of poly-L-lysine(PLL) and laminin were printed on glass coverslips using microcontact printing(μCP) and hippocampal neurons were cultured. We found that neuritogenesis, neurite outgrowth from the soma, preferentially initiated at the vertices of micropatterns. F-actin which is one of key protein for generating neurites tended to concentrate at the vertices of micropatterns, so the vertex preference of neuritogenesis might be a following result of the F-actin concentration at the vertices. Also a major neurite which highly becomes an axon preferentially protrude from the vertex corner of long isosceles triangle patterns. This study was the first attempt to control the direction of neuritogenesis and axonal protrusion using micropatterns which could be easily applied to platforms for analyzing neuronal networks such as multielectrode arrays(MEAs).
배양 기판 표면에 인쇄된 다양한 미세패턴은 체외에서 세포의 이동이나 분열 등의 활동을 제어하는데에도 사용되어왔다. 신경 세포는 신경 돌기 생성과 축색 돌기 돌출이라는 고유의 세포 활동이 있는데 이는 신경 회로망의 연결성을 결정하는데 중요한 역할을 한다. 본 연구에서는 이러한 신경 세포의 활동을 제어하기 위한 미세패턴을 설계하였다. 신경세포체를 제어하기위하여 10 종류의 미세패턴이 130 $μm^2$의 면적으로 제작되었다. 모든 미세패턴은 미세 접촉 프린팅 기술을 이용하여 poly-L-lysine과 laminin을 혼합한 세포흡착분자를 유리 커버슬립 기판에 인쇄하는 방법으로 제작되었으며 해마 신경 세포가 그 위에 배양되었다. 배양된 신경 세포의 신경 돌기 생성은 미세패턴의 꼭지점에서 주로 생성되었다. 이는 신경 돌기를 구성하는 주요 단백질인 F-actin이 꼭지점에 집중되는 경향이 나타나기 때문인 것으로 예상된다. 또한 잠재적 축색 돌기는 긴 이등변 삼각형과 매우 긴 이등변 삼각형 패턴의 꼭지각 쪽에서 생성되는 경향성이 높은 것으로 나타났다. 본 연구는 미세패턴의 형태를 이용하여 신경 돌기 생성과 축색 돌기 돌출을 제어하고자 한 첫 번째 시도이며, 본 연구에서 제시하는 미세패턴은 신경 네트워크 분석 도구인 다채널전극칩에 쉽게 응용 가능하기 때문에 추후 체외 신경 네트워크 연구에 응용될 수 있을 것으로 기대한다.
