Human embryonic stem cells (hESCs) are cells which are capable of self-renewal and can be differentiated into all the cell types in the body, and this makes the hESCs to make use in stem cell therapy, drug discovery, and developmental studies. To utilize the hESCs, it is important to discover differentiation mechanisms and method for efficient differentiation. There are many possible regulation mechanisms on differentiation, and microRNAs are highlighted in these days.
MicroRNAs regulate gene expression by post-transcription mechanism, and regulate mRNA translation in animal cells, and play key roles in cellular differentiation. Like mRNAs, microRNAs are transcribed by RNA polymerase II, and processed by Drosha and Dicer. Mature microRNAs bind to 3’UTR of target mRNA and repress translation.
This study demonstrates the differential expression pattern of microRNAs and mRNAs during the differentiation of hESCs into hESC-derived definitive endoderm (DE) cells, DE cells-derived hepatocytes, hESC-derived CD34 positive cells, and CD34 positive cells-derived endothelial cells. The two lineages show differential expressions of mRNAs and microRNAs related to cytoskeleton and cell-adhesion. Endothelial cells make clustered cells, such as hepatocytes and gut-lining cells, and CD34 positive cells make dispersed cells such as blood cells. In this view, the different expression pattern of mRNAs and microRNAs seems biologically relevant.
Among microRNAs which show dramatic changes during hepatic differentiation, miR-21, miR-214, and miR-216a which are negative regulators of PTEN were enriched in hESC-derived hepatocytes. PI3K pathway is activated by extracellular signal, and PI3K phosphorylates $PIP_2$ into $PIP_3$, and $PIP_3$ recruits many proteins which has PH domain to the membrane, and transmits signal to the downstream. PTEN de-phosphorylates $PIP_3$ to $PIP_2$, antagonizing the entire signaling pathway. As mRNAs related to focal adhesion and cytoskeleton are highly expressed in hepatocyte lineage, as PI3K pathway is a key regulator in these cell functions, and as many growth factors are required for the hepatocyte differentiation, it is speculative that PI3K pathway plays an important role in the differentiation process.
To see the involvement of PI3K pathway in hepatocyte differentiation, I treated a PI3K inhibitor, Ly294002 to DE cells. As expected, cell morphology and gene expressions of hepatocyte marker genes were remarkably different from the control cells. These results imply that PI3K pathway is involved in hepatocyte differentiation and these microRNAs may regulate this process indirectly. With mRNA and microRNA array of differentiating hESCs, I was able to have a deeper insight of differentiation mechanism, and further study will enable to have even deeper insight.
인간 배아 줄기 세포란, 자가재생산이 가능하고 여러 가지 세포로 분화할 수 있는 가능성이 있는 세포를 말한다. 인간 배아 줄기 세포는 신체 내 모든 기관으로 분화할 수 있기에, 이식 치료법이나 의약품 개발, 그리고 발생 연구에 이용할 수 있다. 이렇게 인간 배아 줄기 세포를 이용하기 위해서는 그 분화 조절 기전과 효율적 분화 유도 방법을 알아내는 것이 중요하다. 여러 가지 발생 분화 조절 기전 중, 최근 마이크로RNA에 의한 분화 조절의 연구가 활발하게 진행되고 있다. 마이크로RNA는 mRNA의 발현을 제어하는 전사-후-조절 방법 중의 하나로, 일반적인 동물 세포의 경우 번역 과정을 억제하는 것으로 알려져 있고, 발생 단계에서도 여러 가지 기능을 담당하고 있다. 마이크로RNA의 발현은 일반적인 mRNA와 마찬가지로, RNA 중합 효소 II 에 의해 전사가 되는데, 이후 Drosha와 Dicer에 의해 완성된 모습의 microRNA, 혹은 miRNA가 된다. 완성된 miRNA는 목표로 하는 mRNA의 3’UTR 부분에 RISC와 함께 붙어 번역 과정을 저해하여 유전자 발현을 조절한다.
인간 배아 줄기 세포가 분화할 수 있는 여러 가지 세포들 중, 본 연구는 CD34 양성 세포와 이 세포에서 유래한 혈관 내피 세포, 내배엽 세포와 이 세포에서 유래한 간세포 분화 과정 중 변화하는 mRNA와 마이크로RNA 의 발현 양상 분석, 그리고 이들과 분화 메커니즘 사이의 상관 관계에 관하여 알아보고 있다. 두 계통간 다르게 발현하는 유전자들과 마이크로RNA들을 보면 전체적으로 세포골격과 세포 부착과 관련된 유전자들이 다르게 발현하고 있는 것을 볼 수 있다. 내배엽 세포의 경우 후에 소화기와 간세포와 같은 밀집된 세포를 만드는데 반하여, CD34 양성 세포의 경우 후에 혈액세포와 같이 분산된 세포를 만들게 된다. 이런 분화 과정으로부터 유추해 볼 때, 이러한 유전자와 마이크로RNA 발현 양상의 차이가 생물학적으로 의의가 있다고 할 수 있겠다.
분화 이후 변화를 보이는 여러 가지 마이크로RNA 중, 간세포 분화과정에서 발현이 많이 증가하는 miR-21, miR-214, 그리고 miR-216a의 경우 모두 PTEN이라는 유전자 발현을 조절하는 것으로 알려져 있다. PI3K 신호 전달 체계는 외부 신호 물질을 받아 활성화되어, $PIP_2$를 $PIP_3$로 바꾸고, $PIP_3$에 붙을 수 있는 PH 도메인을 가진 여러 단백질이 위치를 옮겨 신호를 전달한다. PTEN은 이 중 $PIP_3$를 $PIP_2$로 되돌려, 전체적인 신호를 억제하는 역할을 한다. 위의 마이크로RNA들의 경우 모두 PTEN을 억제하여, 결과적으로 PI3K 신호 전달 체계를 활성화시킬 수 있다. 또한, 앞서 본 유전자와 마이크로RNA의 계통특이적 발현 양상과 연관하여, PI3K 신호 전달 체계는 세포골격의 유지와 부착에 매우 중요한 역할을 담당하고 있다. 그리고 간세포 분화 과정에 있어 여러 가지 성장 인자들이 관여하는데, 일반적으로 PI3K 신호 전달 체계가 성장 인자에 의해 활성화 사실을 볼 때 따라서, 간세포 분화 과정에 PI3K 신호 전달 체계의 활성화가 중요할 수 있다는 사실을 마이크로RNA의 발현 결과 예상할 수 있다. PI3K의 조절을 통해, 위의 마이크로RNA들이 간세포 분화를 조절에 관여하는지의 여부를 확인하기 위해, PI3K의 저해제인 LY294002를 분화하는 세포에 처리, 간세포 분화가 저해되는지를 실험해 보았다. 대조군 세포의 경우 전체적으로 덩어리지고, 많은 수의 내포가 형성되어 그물처럼 보이는 모습의 간세포로 분화하였으나, LY294002 처리한 세포의 경우 얇고 넓적한 모양을 가진 세포들로 분화하였다. 간세포 특이적 발현을 보이는 유전자들을 RT-PCR 방법으로 확인한 결과, 처리군 세포는 간세포 유전자를 발현하지 않았다. 이 결과로 볼 때, PI3K 신호 전달이 간세포 분화에 중요하며, 마이크로RNA들이 이 과정에 간접적으로 관여할 수 있는 가능성을 보여주고 있다.
유전자와 마이크로RNA 발현 array을 통하여 인간 배아 줄기 세포의 분화에 대해서 연구할 수 있으며, 앞으로 이들의 더 깊은 분석을 통해 더 자세한 분화 기전 연구를 할 수 있을 것이다.
