Ginseng saponin, the most important secondary metabolite of ginseng, has various pharmacological activities. To obtain minor ginsenoside from major ginsenoside by using microbial biotransformation, a great number of soil bacteria which have strong β ??glucosidase activity were screened. Through four round screenings, 153 strains were found to be able to transform major ginsenosides into minor ginsenosides. One bacterial strain QM49 could convert ginsenosides $Rb_1$, $Rb_2$, Rc, Rd to the active metabolites C-K via F2, C-Y, C-Mc, respectively. Phylogenetically, it was found to belong to genus Mucilaginibacter and was very closely related with Mucilaginibacter kameinonensis $SCK^T$ (97.5% similarity based on 16S rRNA sequence). Enzymatic production of C-K occurred by consecutive hydrolyses of the terminal glucopyranosyl, arabinopyranosyl, arabinofuranosyl moieties at the C-20 carbon and hydrolyses of the terminal and inner C-3 carbon of ginsenosides $Rb_1$, $Rb_2$, Rc showing the biotransformation pathway: $Rb_1$ → Rd → F2 → C-K; $Rb_2$ → C-O → C-Y → C-K; Rc → C-$Mc_1$ → C-Mc → C-K, respectively. The rare minor ginsenoside, C-Mc1, C-Mc, C-O, C-Y were identified by NMR spectrometry and HPLC/MS/MS system.
Two new β-glucosidases from a novel strain of Mucilaginibacter kaistensis ($QM49^T$) obtained from the soil of the field of KAIST (Korea Advanced Institute of Science and Technology) were cloned and characterized. One gene was designated as bgl QM49-1 (2346bp, 781 aa) and the other was designated as bgl QM49-2 (2385 bp, 795 aa). Both enzymes share a low level of similarity which was not aligned easily. However, the function of both enzymes converting ginsenoside was not differentiated too much. Both enzymes catalyzed the conversion of ginsenoside $Rb_1$ to the more pharmacologically active ginsenosides $Rg_3$(S) via Rd and $Rg_1$ to $Rh_1$(S). A BLAST search of the sequence of bgl QM49-1 and bgl QM49-2 revealed significant homology to family 3 glycoside hydrolases. The enzymes catalyzed the hydrolysis of the two glucose moieties attached to the C-20 position of ginsenoside $Rb_1$ and outer glucose attached to the C-20 position $Rg_1$ at pH 8.0 and 30°C. These results indicated that bgl QM49-1 and bgl QM49-2 selectively converts ginsenoside $Rg_1$ to the rare ginsenoside $Rh_1$(S). Herein is the first report of the cloning and characterization of a novel ginsenoside-transforming β-glucosidase of the glycoside hydrolase family 3 which convert the ginsenoside $Rg_1$ to $Rh_1$(S).
Finally, strain $QM49^T$ was classified by using a polyphasic approach. Cells of strain $QM49^T$ were Gram-negative, aerobic, non-spore-forming, non-motile, rod-shaped bacterium. On the basis of 16S rRNA gene sequence similarity, strain $QM49^T$ was shown to belong to the family Sphingobacteriaceae, class Sphingobacteria, and was related most closely to the type strains of Mucilaginibacter kameinonensis $SCK^T$ (97.5% similarity), Mucilaginibacter oryzae $B9^T$ (97.2%), and Mucilaginibacter ximonensis $XM-003^T$ (95.8%). Level of 16S rRNA gene sequence similarity between $QM49^T$ and the type strains of other species in the family Sphingobacteriaceae was less than 95.2%. The G+C content of the genomic DNA of strain $QM49^T$ was 45.2 mol%. Phenotypic and chemotaxonomic data (MK-7 as the major ubiquinone; $iso-C_{15 : 0}, C_{16:1} ω7c/C_{16:1}$ ω as the major fatty acids) supported the affiliation of strain $QM49^T$ to the genus Mucilaginibacter. The results of physiological and biochemical tests allowed genotypic and phenotypic differentiation of strain $QM49^T$ from recognized Mucilaginibacter species. $QM49^T$ therefore considered to represent a novel species of the genus Mucilaginibacter, for which the name Mucilaginibacter kaistensis sp. nov. is proposed. The type strain is $QM49^T$ (= $KACC 14958^T$ = $LMG 25806^T$).
인삼사포닌인 진세노사이드는 인삼의 가장 중요한 이차 대사산물로써 여러가지 약리효과를 보인다. 미생물학적 생물전환법을 이용하여 메이저 진세노사이드로부터 마이너 진세노사이드를 얻기 위해서 수많은 종류의 토양유래 미생물들의 베타-글루코시다제 활성여부를 스크리닝하였다. 베타-글루코시다제 활성이 있는 균주들은 추가적인 진세노사이드 전환능력 실험을 통하여 마지막으로 153 균주가 스크리닝되었다. 이 중 균주 QM49는 진세노사이드 $Rb_1$, Rd, $Rb_2$, Rc, 를 마이너 진세노사이드 F2를 거쳐 C-K로, C-Y, C-Mc로 각각 전환하는 능력을 보여주었다. 계통학적으로 균주 QM49는 뮤시라지니박터 속에 속했고 특히 뮤시라지니박터 카메이노넨시스 $SCK^T$ 균주랑 16S rRNA 유전자 상동성이 97.5%였다. QM49을 10L 배양하여 파쇄 후 조효소를 획득 후 메이저 진세노사이들과 반응시켜 중간 대사체들인 진세노사이드 C-Mc1, C-Mc, C-O, C-Y들인 것을 NMR 분광법과, HPLC/MS/MS 분석법에 의해서 확인할 수 있었다.
KAIST 야산의 토양으로부터 분리된 균주 뮤시라지니박터 카이스텐시스 $QM49^T$로부터 2개의 베타-글루코시다제가 클론되었고 그 특성 연구가 진행되었다. bgl QM49-1 로 명명된 첫번째 유전자 (2346bp, 781 aa)와 bgl QM49-2로 명명된 두 번째 유전자 (2385 bp, 795 aa) 모두 글라이코시다제 패밀리 3 그룹에 속했으나 염기서열 상동성은 매우 낮았다. 비록 두 효소 모두 같은 메커니즘으로 메이저 진세노사이드를 전환하였지만, Bgl QM49-1 의 발현 및 전환 활성은 훨씬 뛰어났다. Bgl QM49-1은 메이저 진세노사이드 $Rb_1$ 를 Rd를 거쳐 $Rg_3$(S) 로, $Rg_1$을 $Rh_1$(S)로 전환시켰다. 이 보고는 글라코시다제 패밀리 3그룹의 진세노사이드 $Rg_1$ 을 전환하는 신규 효소를 클로닝하고 특성연구한 최초의 리포트이다.
마지막으로 균주 $QM49^T$ 는 다상분류법에 의해서 분류되었다. 균주 $QM49^T$의 셀들은 그람 반응 음성, 호기성, 비운동성, 간균 형태였으며 포자 형성은 하지 못하였다. 16S rRNA 유전자 유사성을 비교분석한 결과, 균주 $QM49^T$는 Sphingobacteria 강, Sphingobacteriaceae 과의 Mucilaginibacter (뮤시라지니박터) 속에 속했으며, 가장 가까운 균주로는 Mucilaginibacter kameinonensis $SCK^T$ (97.5% 상동성), Mucilaginibacter oryzae $B9^T$ (97.2%)와 Mucilaginibacter ximonensis $XM-003^T$ (95.8%) 였다. 그 이외의 대표균주들과는 95.2% 이하의 상동성을 보여주었다. 균주 $QM49^T$의 유전체 G+C 비율은 44.6% 였다. 표현형적 데이타, 화학분석 데이타 (MK-7 메이저 퀴논; $iso-C_{15 : 0}, C_{16:1} ω7c/C_{16:1}$ ω 메이저 지방산)들은 균주 $QM49^T$가 Mucilaginibacter속으로 분류된다는 것을 지지해주었다. 생리, 생화적 테스트 결과들은 $QM49^T$가 같은 속 다른 종들과 표현형적, 유전학적으로 구분되어진다는 것을 보여주었다. 그래서, 균주 $QM49^T$는 Mucilaginibacter속의 새로운 종을 대표할 수 있고 그 이름을 Mucilaginibacter kaistensis 로 명명하기를 제안하는 바이다. 그 대표균주는 $QM49^T$ 이며, 다음 두 기관(= $KACC 14958^T$ = $LMG 25806^T$)에 기탁되었다.
