When analyzing resting state brain networks using BOLD (Blood Oxygen Level Dependant) signals, region of interest (ROI) based analysis has become increasing popular in fMRI studies. However one problem in this is the lack of consistent methodology from study to study. In this study, by analyzing a functional network based on resting state and task-induced BOLD signal I identified and established and set of consistent parameters for ROI analysis, and used these methods to quantify functional connectivity between brain structures for specific function and analyzed differences between the left and right hemisphere. I found that major anatomical regions of the cortex have strong functional connectivity during the resting state and that small networks within the motor and somatosensory cortex can be mapped using ROI correlation analysis. Studies in AD progression reveal similar result in deterioration of the Default Mode Network (DMN) with onset of dementia. Also hemispheric analysis suggests that the left hemisphere is more susceptible to AD pathology. Finally I discovered specific regions showing increase in connectivity with AD progression which can possibly be used as a biomarker for AD onset.
BOLD (Blood Oxygen Level Dependent; 혈중 산소 의존) 신호를 이용해 휴지기 뇌 네트워크를 분석할 때, 특정 영역에 기반한 분석은 fMRI 연구에 있어 활발하게 사용되고 있다. 하지만 정립된 분석 방법론의 부재로 인해 다른 연구 그룹 간의 결과 차이가 문제이다. 본 연구에서는 휴지기와 특정 과제 상황에서 나타나는 기능적 네트워크 분석을 통해 영역 분석에 있어 일관성 있는 파라미터를 찾았으며, 이 방법을 사용해 특정 기능에서 나타나는 뇌 구조간의 기능적 연결성을 정량화하고, 좌뇌와 우뇌의 차이를 분석했다. 또한 대뇌 피질의 중요한 구조들이 휴지기 상황에서 강한 기능적 연결성을 보이고 있음을 확인했고 운동, 체지각 피질 내의 작은 네트워크가 영역간의 상관관계 분석을 통해 매핑됨을 확인하였다. 알쯔하이머 병 진행에 관한 연구 역시 비슷하게 치매 발생 시점에서 휴지기 네트워크가 깨어짐을 밝혔다. 마지막으로 알쯔하이머 병이 진행되면서 연결성이 증가하는 특정 부분들을 찾아냈고, 이는 알쯔하이머 발병의 진단 표지자로 활용할 수 있을 것으로 기대한다.
